Pulmonary Alveolar Proteinosis and Multiple Infectious Diseases in a Child with Autosomal Recessive Complete IRF8 Deficiency

Summary: This study describes a case of AR complete IRF8 deficiency in an Argentinian child with multiple infectious diseases and severe pulmonary alveolar proteinosis (PAP). Whole-exome sequencing revealed compound heterozygous variants in the IRF8 gene, and functional analysis confirmed the loss-of-function of both variants. The patient had no blood myeloid mononuclear cells and dendritic cells, suggesting a defective development or function of alveolar macrophages.
