Dissolution Challenges Associated with the Surface pH of Drug Particles: Integration into Mechanistic Oral Absorption Modeling

Summary: This study aimed to predict the systemic exposure of ibuprofen by differentiating its in vitro dissolution profiles, and explore its impact on bioequivalence criteria. In vitro dissolution profiles of ibuprofen obtained under different buffered media were used as input for simulation, and the results matched well with the actual data. In addition, the impact of dissolution rate on bioequivalence was investigated through population simulation.

Prediction of plasma profiles of a weakly basic drug after administration of omeprazole using PBPK modeling

The selection of suitable pH modifiers can improve the dissolution rate and extent of monobasic and dibasic drug compounds under high-gastric pH conditions.
