KRAS: A Druggable Target in Colon Cancer Patients

Francesca Negri et al.

Summary: Mutations in KRAS are common in cancer, but direct targeting of KRAS has been challenging due to its resistance to small molecule inhibition and the design difficulties in creating specific drugs. However, covalent inhibitors of the KRASG12C variant have shown promise and could provide a therapeutic approach for KRAS-driven tumors.

Evaluation of stromal cell infiltration in the tumor microenvironment enable prediction of treatment sensitivity and prognosis in colon cancer

Rui Zhou et al.

Summary: Current clinical factors for screening candidates for adjuvant chemotherapy in colon cancer are inadequate. A novel 31-gene signature called stromal cell infiltration intensity score (SIIS) has been established using machine learning to distinguish patients with high stromal cell infiltration who are at higher risk for recurrence and mortality and may not benefit from chemotherapy. The findings suggest that SIIS provides an important reference for treatment decisions in these special patients.
