Beneficial effects of QTC-4-MeOBnE in an LPS-induced mouse model of depression and cognitive impairments: The role of blood-brain barrier permeability, NF-KB signaling, and microglial activation

Summary: Clinical and preclinical investigations have suggested a biological link between major depressive disorder (MDD) and Alzheimer's disease (AD). A pharmacologic approach using QTC-4-MeOBnE has shown benefits in treating depressive-like behavior and cognitive impairments induced by lipopolysaccharide injections in mice. The effects are associated with restoring blood-brain barrier permeability, reducing oxidative/nitrosative biomarkers, and decreasing neuroinflammation mediated NF-KB signaling.

Summary: In this study, QTC-4-MeOBnE was found to prevent Aβ formation by downregulating APP and BACE levels, and reduce the levels of phosphorylated forms of tau through modulation of the GSK3β pathway in AD cellular models. These findings provide new insights into the mechanism of action of QTC-4-MeOBnE and further support its potential as a therapeutic strategy against AD.
