Downregulation of TREM2 expression exacerbates neuroinflammatory responses through TLR4-mediated MAPK signaling pathway in a transgenic mouse model of Alzheimer's disease

Xiaoqian Peng et al.

Summary: Activation of glial cells and neuroinflammation are important in Alzheimer's disease (AD). This study shows that downregulation of the microglial receptor TREM2 exacerbates AD-related neuropathology and triggers neuroinflammatory responses through the TLR4-MAPK pathway.

Tanshinone IIA regulates glycogen synthase kinase-3β-related signaling pathway and ameliorates memory impairment in APP/PS1 transgenic mice

Xiaoqian Peng et al.

Summary: This study demonstrates that tanshinone IIA (tan IIA) can ameliorate cognitive decline and tau pathology in APP/PS1 mice by modulating the PI3K/Akt/GSK-3β signaling pathway.

TREM2 overexpression rescues cognitive deficits in APP/PS1 transgenic mice by reducing neuroinflammation via the JAK/STAT/SOCS signaling pathway

John Bosco Ruganzu et al.

Summary: The overexpression of TREM2 was found to rescue cognitive deficits, reduce Aβ plaque deposition, decrease synaptic and neuronal loss, as well as ameliorate neuroinflammation, likely through the inhibition of neuroinflammatory responses via the JAK/STAT/SOCS signaling pathway and promotion of M2 microglia activation. This study highlights TREM2 as a potential target against AD by shifting microglia from an M1 to M2 phenotype.

A current view on Tau protein phosphorylation in Alzheimer's disease

Susanne Wegmann et al.

Summary: The functions of Tau in the central nervous system are regulated by various posttranslational modifications, with phosphorylation being widely recognized as a pathological hallmark in diseases like AD. However, the similarities and heterogeneity of phosphorylation patterns between physiological and pathological Tau make it challenging to identify specific modifications as therapeutic targets and biomarkers.

Clinical diagnosis of Alzheimer's disease: recommendations of the International Working Group

Summary: The use of biomarkers in the diagnosis of Alzheimer's disease has limitations and should be used cautiously in a clinical setting, restricted to individuals with specific phenotypes and positive biomarkers. Biomarker-positive cognitively unimpaired individuals should be considered at-risk for progression to Alzheimer's disease.
