Effects of triheptanoin (UX007) in patients with long-chain fatty acid oxidation disorders: Results from anopen-label,long-termextension study

Jerry Vockley et al.

Summary: Triheptanoin has been shown to reduce the rate and duration of major clinical events in patients with LC-FAOD, with the most common related adverse events being mild to moderate. Safety profile is consistent with previous observations.

Impact of newborn screening on the reported incidence and clinical outcomes associated with medium- and long-chain fatty acid oxidation disorders

Deborah Marsden et al.

Summary: Fatty acid oxidation disorders are potentially fatal inherited disorders that can be managed through early disease detection and dietary intervention. Newborn screening plays a crucial role in prompt identification and management of patients, leading to improved outcomes with dietary management.

Different Lipid Signature in Fibroblasts of Long-Chain Fatty Acid Oxidation Disorders

Khaled I. Alatibi et al.

Summary: Research has shown significant changes in cellular lipidome in patients with long-chain fatty acid oxidation disorders, including remodeling of mitochondrial cardiolipins, abnormal phosphatidylcholine/phosphatidylethanolamine ratio, and increased content of plasmalogens, supporting an inflammatory phenotype. In addition, LCHAD deficiency patients exhibit increased ratios of sphingomyelin/ceramide and sphingomyelin/hexosylceramide, which may be related to the neuropathic phenotype of LCHADD/mitochondrial trifunctional protein deficiency.

Development and characterization of a mouse model for Acad9 deficiency

Andrew Sinsheimer et al.

Summary: ACAD9 is an enzyme involved in mitochondrial fatty acid oxidation, and its deficiency leads to combined defects in fatty acid oxidation and oxidative phosphorylation. Animal models of ACAD9 deficiency may serve as useful tools for developing novel therapeutics.

Pervasive inflammatory activation in patients with deficiency in very-long-chain acyl-coA dehydrogenase (VLCADD)

Abbe N. Vallejo et al.

Summary: The study found that patients with VLCADD exhibit systemic inflammatory responses during symptomatic and asymptomatic periods, with an abundance of immune cells expressing high levels of inflammatory mediators. Patients with recurrent hospitalisations showed sustained high levels of plasma cytokines and higher intracellular stores of inflammatory molecules in monocytes.
