MicroRNA-10a-5p targets SERPINE1 to suppress cell progression and epithelial-mesenchymal transition process in clear cell renal cell carcinoma

This study found that miR-10a-5p plays a crucial role in the progression of clear cell renal cell carcinoma (ccRCC) by inhibiting the expression of SERPINE1. It can restrain the proliferative, migratory, and invasive abilities of ccRCC cells and promote cell apoptosis. These findings provide a new approach for the treatment of ccRCC.
Background Clear cell renal cell carcinoma (ccRCC) is a serious threat to human life and health. The existing molecular targeted therapy of ccRCC has encountered the problem of drug resistance, so it is necessary to explore new therapeutic targets. Some publications have shown that miR-10a-5p plays a crucial role in regulating various cancers, but few articles have investigated the regulation of miR-10a-5p in the progression of ccRCC.Objective This study aimed to probe into the impacts of miR-10a-5p on ccRCC cell phenotype progression.Results Bioinformatics analysis showed that the expression level of miR-10a-5p was downregulated in ccRCC tissues and cell lines. The results of the cellular experiments demonstrated that miR-10a-5p constrained proliferative, migratory and invasive abilities of ccRCC cells, and facilitated cell apoptosis. Dual-luciferase assay confirmed the targeting relationship between miR-10a-5p and SERPINE1. Besides, loss-of-function studies showed that SERPINE1 facilitated the malignant progression and epithelial-mesenchymal transition (EMT) of ccRCC. Rescue experiments stated that miR-10a-5p partially repressed ccRCC progression and EMT process by targeting SERPINE1.Conclusion Collectively, our results indicated that miR-10a-5p is expected to be a potential target for ccRCC, providing a new approach for ccRCC treatment.
