Pierce into Structural Changes of Interactions Between Mutated Spike Glycoproteins and ACE2 to Evaluate Its Potential Biological and Therapeutic Consequences

Summary: This study examines the effects of mutations in the spike protein of SARS-CoV-2 on its structural consequences and therapeutic efficacy. By predicting the 3D structure of mutant spike proteins and analyzing their interactions with ACE2 proteins and an engineered therapeutic ACE2, the study finds that mutations alter the interaction orientation and increase the affinity between the spike protein and ACE2. Additionally, mutations also impact the cavity size and residue depth. The study concludes that mutated spike proteins demonstrate higher binding affinity and designed ACE2-based therapeutics remain highly effective against COVID-19.
