Enhancing Soluble Expression of Phospholipase B for Efficient Catalytic Synthesis of L-Alpha-Glycerylphosphorylcholine

Summary: This study successfully obtained high-activity phospholipase B (PLB) and established an efficient system for L-alpha-glycerylphosphorylcholine (L-alpha-GPC) synthesis. The highest catalytic activity of PLB was achieved through the co-expression with DnaKJE. By optimizing the expression conditions, an optimal PLB activity was achieved. Using organic-aqueous mixed systems and surfactant-supplemented aqueous systems, a high yield of L-alpha-GPC was obtained. The proposed cost-effective PLB production and environmentally friendly PLB-catalyzed system offer a candidate strategy for the industrial production of L-alpha-GPC.

Summary: Through genetic engineering, Halomonas bluephagenesis with high tolerance successfully produces high titers of itaconic acid (IA) from citrate using a cell catalytic strategy. Under optimized conditions, the conversion rate reaches 93.24% with a productivity of 1.63 g/(L h).

Synthetic Protein Scaffolds for Improving R-(-)-Linalool Production in Escherichia coli

Summary: By physically colocalizing three enzymes in synthetic complexes, the production efficiency of R-(-)-linalool in Escherichia coli was successfully improved. Through further optimization of inducer, temperature, and glycerol concentration, the production titer in the shake flask increased by approximately 10 times compared to the scaffold-free control, with a significant increase in production yield in the fermenter as well.
