Summary: The expression of immune checkpoint PD-L1 plays a crucial role in maintaining an immunosuppressive microenvironment, regulating metabolism and signaling pathways in clear cell renal cell carcinoma (ccRCC). PD-L1 is required for mediating some of the effects of IFN gamma in ccRCC cells, highlighting its importance in regulating the metabolism of ccRCC cells in response to inflammatory signals.

Summary: Our study demonstrates that CD8(+) tissue resident memory T (T-RM) cells are the main activated T cell subset in ICI colitis. The pattern of gastrointestinal immunopathology in ICI colitis is distinct from ulcerative colitis at both the immune and epithelial-signaling levels. Activation of CD8+ T-RM cells correlates with clinical and endoscopic severity of ICI colitis.

Next generation of anti-PD-L1 Atezolizumab with enhanced anti-tumor efficacy in vivo

Summary: Avelumab, an IgG1 anti-PD-L1 monoclonal antibody, has been approved for certain cancers but its rapid clearance and shorter half-life compared to other antibodies is not fully understood. Our study suggests that the unique internalization mechanism of avelumab, involving both Fc gamma R and PD-L1 binding, may explain its fast clearance and provides insights for future therapeutic development.
