Mass spectrometry-based strategies for single-cell metabolomics

Rui Hu et al.

Summary: Single cell analysis is of increasing interest to the research community due to its ability to analyze cellular heterogeneity and discover new biomarkers. With advancements in technology, it is now possible to perform multiple omics at the single cell level. However, single-cell metabolomics poses unique challenges due to complex compound composition and the inability to amplify metabolites. Despite these challenges, significant progress has been made in mass spectrometry-based single-cell metabolomics.

Liquid-state NMR spectroscopy for complex carbohydrate structural analysis: A hitchhiker's guide

Immacolata Speciale et al.

Summary: This article provides a practical guide to the structural determination of carbohydrates using liquid-state NMR. The narrow range of chemical shifts in the NMR signals of carbohydrates makes it challenging to identify individual monosaccharide units. However, attributing the NMR chemical shifts can reveal the nature of the constituents, their interconnections, and even their absolute configuration, paving the way for more sophisticated analyses.

Synergies between Hyperpolarized NMR and Microfluidics: A Review

James Eills et al.

Summary: Hyperpolarized nuclear magnetic resonance (NMR) and lab-on-a-chip microfluidic technology are two distinct fields of research that have recently shown increasing overlap. Microfluidic integration allows for complex experimental steps to be combined onto a single platform, making it more widely applicable in medical diagnostics, forensic analysis, and biomedical research. Nuclear magnetic resonance is a high-resolution spectroscopic technique that can monitor biological and chemical processes in real-time, but its low sensitivity has limited its use. Recent advances in hyperpolarization techniques have the potential to enhance NMR signals by up to 5 orders of magnitude, expanding its utility in microfluidic systems. The implementation of microfluidic technology can benefit the complex chemical and physical manipulations required for hyperpolarization. This review examines the intersection between hyperpolarization-enhanced NMR and microfluidics, and assesses how these fields have mutually benefited each other.

Single-Cell Profiling of Fatty Acid Uptake Using Surface-Immobilized Dendrimers

Zhili Guo et al.

Summary: This study presents a chemical approach to analyze fatty acid uptake in single cells, which involved using azide-modified analogues and surface-immobilized dendrimers with DBCO groups. By integrating this method into a microfluidics-based multiplex protein analysis platform, the researchers were able to explore the relationships between fatty acid influx, oncogenic signaling activities, and cell proliferation. The study revealed differential correlations between p70S6K and 4EBP1 with fatty acid uptake, and demonstrated synergistic inhibition of cell proliferation through co-targeting p70S6K and fatty acid metabolism. This work provided new insights into cancer biology by studying fatty acid metabolism in the context of protein signaling at single-cell resolution.
