Genetic pain loss disorders

Annette Lischka et al.

Summary: This article discusses the epidemiology, pathophysiology, diagnosis and treatment of genetic pain loss disorders, which are characterized by reduced pain sensation. The development of these disorders is associated with recurrent injuries, burns and poorly healing wounds. Current treatment options mainly focus on symptomatic relief, but targeted therapies are being explored. The understanding of the genes and mechanisms related to pain loss disorders also provides new insights for the development of innovative pain medicines.

A central mechanism of analgesia in mice and humans lacking the sodium channel NaV1.7

Donald Iain MacDonald et al.

Summary: Deletion of SCN9A encoding Na(V)1.7 in humans leads to pain insensitivity and anosmia. Knockout mice show normal nociceptor activity, but synaptic transmission from nociceptor terminals in the spinal cord is reduced by an opioid-dependent mechanism, reversing analgesia. This mechanism is also responsible for anosmia.

Pain behavior in SCN9A (Nav1.7) and SCN10A (Nav1.8) mutant rodent models

Yaping Xue et al.

Summary: The review summarizes the findings from rodent models with Scn9a and Scn10a gene mutations, showing that loss-of-function or knockout of these genes decrease pain sensitivity, while certain mutations can lead to painful small fiber neuropathy. Future research on these mutations in preclinical models can provide insights into the impact of these mutations on pain.

Hereditary sensory and autonomic neuropathy in a family of mixed breed dogs associated with a novel RETREG1 variant

Rodrigo Gutierrez-Quintana et al.

Summary: Hereditary sensory and autonomic neuropathies (HSANs) are genetic disorders affecting the peripheral nervous system. A study on a mixed breed dog family confirmed a disorder affecting sensory and autonomic nerves, and identified a novel and potentially pathogenic RETREG1 variant through whole genome sequencing.
