Multi-method global sensitivity analysis of mathematical models

An Dela et al.

Summary: This paper presents a multi-method framework that combines three global sensitivity analysis methods. The algorithms are explained with MATLAB codes and guidelines for tuning hyper-parameters. Two example models are used to demonstrate the methodology and workflow, and a graphics tool is provided for comparing the results of the sensitivity analysis algorithms.

Mathematical Modeling of Vaccines That Prevent SARS-CoV-2 Transmission

David A. Swan et al.

Summary: Clinical trials of SARS-CoV-2 vaccines assess efficacy against symptomatic disease, but may not fully differentiate between preventing infection or preventing symptoms. Using mathematical modeling, it is shown that high vaccine efficacy not only requires preventing transmission after infection, but also minimizing symptoms post-infection. Different vaccines may achieve high efficacy through varying mechanisms, with vaccines that offer complete protection against infection or transmission reducing the number of vaccinations needed for herd immunity.

Neutralizing antibody levels are highly predictive of immune protection from symptomatic SARS-CoV-2 infection

David S. Khoury et al.

Summary: The level of neutralizing antibodies is closely related to immune protection against COVID-19, playing a crucial role in protecting against detected infection and severe infection. Studies have shown that neutralizing titers will decline over time after vaccination, leading to decreased protection against SARS-CoV-2 infection.

A quantitative model used to compare within-host SARS-CoV-2, MERS-CoV, and SARS-CoV dynamics provides insights into the pathogenesis and treatment of SARS-CoV-2

Kwang Su Kim et al.

Summary: The study compared the viral dynamics of SARS-CoV-2 with MERS-CoV and SARS-CoV, finding that SARS-CoV-2 had a higher within-host reproduction number and a shorter time to viral load peak. Therapies targeting de novo infection or virus production were found to be effective if initiated before the viral load peak, while therapies promoting cytotoxicity of infected cells were less sensitive to treatment timing. Combining both types of therapies showed synergistic effects in reducing viral load.

A Mathematical Model of COVID-19 with Vaccination and Treatment

M. L. Diagne et al.

Summary: A mathematical model was developed to analyze the COVID-19 transmission mechanism and the impact of therapeutic measures, showing globally stable disease-free and endemic equilibriums when R-0(v) is less or greater than unity. Findings also revealed the critical vaccination threshold dependent on vaccine efficacy and highlighted the most impactful parameters through sensitivity analysis.
