Serum neurofilament light chain for individual prognostication of disease activity in people with multiple sclerosis: a retrospective modelling and validation study

Pascal Benkert et al.

Summary: sNfL is a biomarker of neuronal damage used in monitoring disease activity in multiple sclerosis patients. This study established a reference database to correct for age and BMI, showing sNfL's potential in identifying individuals at risk for future disease activity. The results suggest that sNfL Z scores could predict acute and chronic disease activity in multiple sclerosis patients, with higher Z scores indicating increased risk.

Neurofilament light chain, a biomarker for polyneuropathy in systemic amyloidosis

Summary: The study found that sNfL levels are increased in patients with PNP in both AL and ATTRv amyloidosis, and are related to the severity of PNP in ATTRv amyloidosis. This suggests that sNfL may be a promising biomarker for detecting PNP.

Neurofilament Light Chain as a Biomarker of Hereditary Transthyretin-Mediated Amyloidosis

Summary: This study identified changes in proteome associated with onset and progression of hATTR amyloidosis, with findings suggesting that NfL may serve as a biomarker of nerve damage and polyneuropathy in ATTRv amyloidosis, enabling earlier diagnosis and disease progression monitoring. Results showed patisiran treatment trending the proteome of patients towards healthy controls and significantly correlating with reduced NfL levels and neuropathy impairment score improvement.

Serum Neurofilament Light Chain Levels in the Intensive Care Unit: Comparison between Severely Ill Patients with and without Coronavirus Disease 2019

Raoul Sutter et al.

Summary: There is increasing evidence for various neurological manifestations of COVID-19, with higher sNfL concentrations observed in critically ill patients with COVID-19 compared to healthy controls and critically ill non-COVID-19 patients. Elevated sNfL levels were associated with unfavorable short-term outcomes, indicating common and pronounced neuronal injury in critically ill patients.
