Controlling the Direction of S-Nitrosation versus Denitrosation: Reversible Cleavage and Formation of an S-N Bond within a Dicopper Center

Wenjie Tao et al.

Summary: The interconversion between metal-S-nitrosothiol adduct and metal nitrosyl thiolate complex can regulate the direction of reversible S-(de)nitrosation. This process is sensitive to temperature, solvent coordination ability, and counterions, and copper centers can modulate the bond formation and cleavage through minor perturbations of the local environment.

Synthesis and characterization of a model complex for flavodiiron NO reductases that stabilizes a diiron mononitrosyl complex

Hai T. Dong et al.

Summary: Flavodiiron NO reductases (FNORs) are important enzymes in microbial pathogenesis, and the second coordination sphere (SCS) hydrogen bond network is critical for the NO reduction reaction catalyzed by FNORs. A model complex of FNORs with pendant hydrogen bond donors was synthesized, and a strategy to stabilize mononitrosyl diiron complexes using the SCS was discovered.

Distortion of the [FeNO]2 Core in Flavodiiron Nitric Oxide Reductase Models Inhibits N-N Bond Formation and Promotes Formation of Unusual Dinitrosyl Iron Complexes: Implications for Catalysis and Reactivity

Corey J. White et al.

Summary: The structure of the active site greatly affects the ability of the diiron core to mediate the formation of nitrous oxide. The absence of a bridging carboxylate in the diiron core leads to expansion, resulting in elongated N-N distances and distorted N-Fe-Fe-N dihedral angles. Compared to the original model compound that quantitatively reduces NO to N2O, these analogues show reduced N2O production and the formation of unprecedented dinitrosyl iron complexes. However, these dinitrosyl iron complexes exhibit unexpected reactivity and can produce significant amounts of N2O after 2 hours.

NO Coupling at Copper to cis-Hyponitrite: N2O Formation via Protonation and H-Atom Transfer

Pokhraj Ghosh et al.

Summary: Copper nitrite reductases convert nitrite ions to nitric oxide and nitrous oxide at high levels of nitrogen oxide flux. The assembly of nitric oxide at a single copper site to form copper cis-hyponitrites has been demonstrated in the presence of an external reductant. Furthermore, the removal of a fraction of the structure in the copper cis-hyponitrite complex can be achieved by adding specific reagents, resulting in the formation of a spectroscopically similar anion. These studies provide insights into the reductive coupling of nitrogen oxides at a single copper center and the utilization of the strong oxidizing power of nitrite.

What Is the Right Level of Activation of a High-Spin {FeNO}7 Complex to Enable Direct N-N Coupling? Mechanistic Insight into Flavodiiron NO Reductases

Hai T. Dong et al.

Summary: In this study, a new model system for flavodiiron nitric oxide reductases (FNORs) was reported, which can mediate direct nitric oxide (NO) reduction or stabilize the first intermediate of the reaction. The results demonstrate that the high-spin {FeNO}7 complexes require a higher activation level for NO reduction compared to those in FNORs, highlighting the importance of hydrogen bonding residues in the active sites of FNORs.

Summary: This study investigates the gas-solid reaction of nitric oxide with the metal-organic framework Cu-I-ZrTpmC* and identifies an unusual chelating N2O2.- intermediate using in situ spectroscopies and density functional theory simulations. The results highlight the advantage of site-isolation in metal-organic frameworks (MOFs) for studying important reaction intermediates and provide a mechanistic scenario compatible with the proposed one-electron couple in these enzymes.

Direct NO Reduction by a Biomimetic Iron(II) Pyrazolate MOF

Zhongzheng Cai et al.

Summary: A novel metal-organic framework has been synthesized, containing Fe2+ chains bridged by dipyrazolate linkers and N,N-dimethylformamide ligands. This MOF facilitates direct reduction of nitric oxide, producing nearly quantitative yields of nitrous oxide and emulating the reactivity of flavodiiron nitric oxide reductases. The ferrous form of the MOF can be regenerated via a synthetic cycle involving reduction with cobaltocene followed by reaction with trimethylsilyl triflate.
