Diversity-Oriented Synthesis and Chemoinformatics: A Fruitful Synergy towards Better Chemical Libraries

Elena Lenci et al.

Summary: In the past two decades, Diversity-Oriented Synthesis (DOS) has undergone significant changes, shifting from a focus on structural diversity to incorporating new approaches to enhance the biological outcomes of molecules. Chemoinformatics has played a crucial role in identifying unexplored biologically-relevant regions of chemical space and visualizing and predicting the structural diversity and complexity of compounds within DOS libraries.

Digitization and validation of a chemical synthesis literature database in the ChemPU

Simon Rohrbacht et al.

Summary: Despite potential, the automation of synthetic chemistry has only made slow progress in recent decades. In this study, we introduce an automated chemical reaction database with 100 representative molecules. Through robotic experimentation, over 50 reactions from the database were performed, achieving yields and purities comparable to those achieved by experts.

Convergence of multiple synthetic paradigms in a universally programmable chemical synthesis machine

Davide Angelone et al.

Summary: The study demonstrates that the Chemputer synthesis robot can be programmed to perform a variety of reactions, including solid-phase peptide synthesis, iterative cross-coupling, and diazirine chemistry. Developing universal and modular hardware that can be automated makes various batch chemistry accessible. The research also showcases a complex convergent robotic synthesis, highlighting the utility of the Chemputer system.
