Catalytic role of carbonyl oxygens and water in selinadiene synthase

Summary: This study reveals the dual role of the main-chain carbonyl oxygen of Gly182 in selina-4(15),7(11)-diene synthase (SdS) as both a base and an acid in terpene synthase (TS) catalysis. It also demonstrates the importance of the G1/2 helix break in TS catalysis, as the Gly182 in this region plays a crucial role in recognizing substrate conformation, stabilizing carbocation intermediates, and anchoring their poses. The conservation of the G1/2 helix break in various organisms suggests its general significance in TS catalysis.

Identification and Characterization of Bifunctional Drimenol Synthases of Marine Bacterial Origin

Ancient plant-like terpene biosynthesis in corals

Summary: Octocorals contribute significantly to the chemical diversity of terpenoids in the ocean. Contrary to the conventional belief that natural products in sessile marine animals are primarily synthesized by symbiotic microbes, this study provides evidence of a monophyletic lineage of animal-encoded terpene cyclases (TCs) present in octocorals. The study identified several TC enzymes in octocorals that produce precursors of specific terpenoids unique to corals. The research highlights the biosynthetic capacity of the Animalia kingdom, comparable to plants, and suggests the biotechnological potential of octocorals for biomedical applications.

Terpene Cyclases and Prenyltransferases: Structures and Mechanisms of Action

Summary: In this study, the structures of sesquisabinene and santalene synthases from Santalum album L. were obtained, revealing the relationship between Mg2+ and catalytically active conformation, as well as the presence of the D-motif in cyclases and prenyltransferases. Additionally, it was found that many highly conserved residues in these proteins are located outside the active site region, suggesting their evolutionary origin from beta gamma domain proteins, while a conserved alpha-domain Arg forms a salt bridge with Glu to stabilize the catalytically active structure.
