A Modular and Expandable Ecosystem for Metabolomics Data Annotation in R

Johannes Rainer et al.

Summary: Liquid chromatography-mass spectrometry (LC-MS)-based untargeted metabolomics experiments require customized annotation workflows. We present an ecosystem of R packages that provide a modular infrastructure for annotating untargeted metabolomics data, including initial annotation and reference library comparison. The system supports various data formats and offers highly customizable functionality for most untargeted LC-MS data.

Iron Dysregulation in Mitochondrial Dysfunction and Alzheimer's Disease

John O. Onukwufor et al.

Summary: Alzheimer's disease is a neurodegenerative disease characterized by neuronal dysfunction and memory and cognitive decline. Iron dysregulation and iron-dependent cell death are associated with AD. Targeting ferroptosis, a specific form of cell death, may be a potential therapeutic intervention for AD.

Novel Extraction Method for Combined Lipid and Metal Speciation From Caenorhabditis elegans With Focus on Iron Redox Status and Lipid Profiling

Bastian Blume et al.

Summary: The progression of Parkinson's disease is linked to iron redox status homeostasis and ROS formation, with lipids being the primary targets of ROS. A novel extraction method using EGTA as a stabilizing agent and synthetic quartz beads has been demonstrated to be faster, easier, and more reliable for determining iron redox status in vivo. Combining metal extraction with lipid extraction protocols using MTBE allows for the simultaneous determination of lipid profiles and metal species, effectively addressing the challenges of lipid precipitation and total metal content determination in frozen samples.

Zinc Affects Cholesterol Oxidation Products and Fatty Acids Composition in Rats' Serum

Agnieszka Stawarska et al.

Summary: The study showed that a diet enriched with zinc has anti-tumor effects in rats with breast cancer, reducing tumor number and size, as well as cholesterol and cholesterol oxidation products levels. Zinc nanoparticles influence fatty acid profile and inhibit desaturating enzyme activity in rats' serum.

Toxic Feedback Loop Involving Iron, Reactive Oxygen Species, alpha-Synuclein and Neuromelanin in Parkinson's Disease and Intervention with Turmeric

Zune Jansen van Rensburg et al.

Summary: Parkinson's disease is a movement disorder caused by severe loss of dopaminergic neurons in the substantia nigra. The disease pathology involves a toxic loop of reactive oxygen species, iron, alpha-synuclein, and neuromelanin, ultimately leading to neuronal death. Targeting the increase in ROS and iron levels may be a potential therapeutic approach to disrupt this toxic cycle and complement existing PD treatments.

Investigation of cardiolipin oxidation products as a new endpoint for oxidative stress in C. elegans by means of online two-dimensional liquid chromatography and high-resolution mass spectrometry

Patrick O. Helmer et al.

Summary: Research on neurodegenerative and age-related diseases highlights the role of oxidative stress, with a focus on the unique lipid class cardiolipin (CL) as a sensitive marker for oxidative stress in diseases like Parkinson's and Alzheimer's. Using C. elegans as a model organism, studies have demonstrated the significance of CL oxidation products as a potential marker for oxidative stress, particularly in relation to mitochondrial function.
