Measuring single-virus fusion kinetics using an assay for nucleic acid exposure

Ana M. Villamil Giraldo et al.

Summary: The kinetics of virus-membrane fusion provides important insights into viral entry mechanisms. Researchers have developed a real-time assay to detect individual virus genome exposure, allowing observation of different stages of viral entry. Fluorescent probes are used to specifically detect fusion events. Results show that the fusion kinetics of influenza virus with liposomes of different sizes are similar, suggesting that the starting curvature of the liposomes does not affect the rate of influenza entry.

Precise Triggering and Chemical Control of Single-Virus Fusion within Endosomes

Sourav Haldar et al.

Summary: The study focused on the chemically controllable triggering of single-virus fusion within endosomes. The results showed that endosomal curvature did not significantly affect fusion kinetics, indicating that membrane deformability may play a more crucial role in viral entry. This study provides insights into how cellular restriction factors and altered endosomal trafficking affect viral membrane fusion.

Single-virus content-mixing assay reveals cholesterol-enhanced influenza membrane fusion efficiency

Katherine N. Liu et al.

Summary: The study introduces a new method to study the content-mixing process of viruses within single cells by binding content-labeled vesicles to unlabeled influenza virus. Results show that the cholesterol content of the target membrane can increase the efficiency of viral content mixing and decrease the proportion of events resulting in content loss. This approach may be promising for studying the single-virus content-mixing kinetics of other enveloped viruses.

The late endosome-resident lipid bis(monoacylglycero)phosphate is a cofactor for Lassa virus fusion

Ruben M. Markosyan et al.

Summary: This study investigated the cell-cell fusion induced by the Old World Lassa virus GPC complex and found that viral entry into host cells is mainly regulated by the human LAMP1 receptor and a specific anionic lipid, bis(monoacylglycero)phosphate. The findings suggest that arenaviruses have evolved to utilize bis(monoacylglycero)phosphate to enter cells from late endosomes.
