Juggling with fluorescent proteins: Spectrum and structural changes of the mCardinal2 variants

Summary: mCardinal2 is a red fluorescent protein developed from the designs of mKate, mNeptune, and mCardinal. The brightness and fluorescence spectrums of mCardinal2 are influenced by its mutants, resulting in different fluorescent colors. Analysis of the crystal structures of the six variants showed that the brightness-increased variant C158D exhibited enhanced hydrogen bonds around the chromophore, while brightness-decreased variants showed diminished hydrogen bonds.

Temperature-Dependent Fluorescence of mPlum Fluorescent Protein from 295 to 20 K

Energetic Basis and Design of Enzyme Function Demonstrated Using GFP, an Excited-State Enzyme

Summary: The past decades have seen a surge in the design of de novo proteins, but it remains challenging to design catalytic functions that can compete with naturally occurring counterparts. Directed evolution often provides efficient solutions, but the fitness landscape is not well understood. Green fluorescent protein (GFP) has been extensively redesigned and, despite not being an enzyme in the conventional sense, it exhibits catalytic behavior similar to conventional ground-state catalysts. In this study, the authors show that designed mutants and hybrids of GFP can provide a quantitative model for predicting the influence of sterics and electrostatics on its catalytic behavior. This research offers important insights into the design of protein function and catalysts.

Opening of glutamate receptor channel to subconductance levels

Directed Evolution of a Bright Variant of mCherry: Suppression of Nonradiative Decay by Fluorescence Lifetime Selections

Summary: This study focuses on the lifetime-based directed evolution of mCherry, a popular red-fluorescent protein (RFP), which has successfully improved its brightness by identifying specific mutations near the chromophore. The study reveals a linear relationship between lifetime and quantum yield, as well as consistent blue-shifts of the absorption and emission spectra. The improvement in brightness is primarily attributed to a decrease in nonradiative decay of the excited state. The findings suggest that nonradiative mechanisms beyond energy-gap models are suppressed in this specific evolution trajectory.

Local Electric Field Controls Fluorescence Quantum Yield of Red and Far-Red Fluorescent Proteins

By evaluating local electric fields experienced by the chromophore in the protein environment, it was found that the twisted intramolecular charge transfer mechanism plays a dominant role in most studied RFPs, providing important guidelines for shifting the absorption wavelength to the red while maintaining high brightness.
