Endocannabinoid-Mediated Control of Neural Circuit Excitability and Epileptic Seizures

Summary: Research on endocannabinoid signaling has provided valuable insights into the control of neural circuit excitability in both healthy and diseased states. Recent advancements in the development of cannabinoid sensors have allowed for a better understanding of the activity-dependent changes in endocannabinoid signaling during seizures. Furthermore, this research has opened up possibilities for the clinical use of cannabidiol in the treatment of refractory epilepsy.

The endocannabinoid system plays a significant role in modulating emotional behavior, and alterations in this system may contribute to the development of behavioral comorbidities in response to peripheral inflammation.

Summary: The endocannabinoid 2-arachidonoylglycerol, not anandamide, is found to be dynamically linked to hippocampal neural activity in awake behaving mice. Additionally, seizures significantly amplify the levels of endocannabinoids and drive the synthesis of vasoactive prostaglandins, culminating in stroke-like events. These findings provide new insights into normal and pathological endocannabinoid signaling in vivo.
