The multi-faceted role of mitochondria in the pathology of Parkinson's disease

Dennison Trinh et al.

Summary: Mitochondria play a crucial role in neuronal function, regulating protein synthesis and synaptic plasticity. Dysfunction of mitochondria has been associated with several neurodegenerative diseases, including Parkinson's disease.

Brief Sensory Deprivation Triggers Cell Type-Specific Structural and Functional Plasticity in Olfactory Bulb Neurons

Elisa Galliano et al.

Summary: Temporary sensory deprivation can induce structural and functional plasticity in dopaminergic neurons in the olfactory bulb, resulting in changes in axon initial segment length and intrinsic excitability. This experience-dependent plasticity may help fine-tune sensory processing in the face of continuously fluctuating inputs.

Intracranial administration of alpha-synuclein fibrils in A30P-synuclein transgenic mice causes robust synucleinopathy and microglial induction

Renee C. Gentzel et al.

Summary: Research has shown that pre-formed fibrils (PFFs) of recombinant human alpha Syn have distinct effects on mice overexpressing human A3OP alpha Syn, inducing regionally-specific accumulations of phosphorylated synuclein and microglial induction. A3OP mice display a different motor phenotype compared to wildtype mice, but PFF-induced loss of tyrosine hydroxylase is similar in both types of mice.

α-Synuclein Spread from Olfactory Bulb Causes Hyposmia, Anxiety, and Memory Loss in BAC-SNCA Mice

Norihito Uemura et al.

Summary: This study aimed to investigate the clinicopathological contribution of alpha-Syn spread from the olfactory bulb in PD. The results showed that alpha-Syn preformed fibril injections induced more widespread pathology in transgenic mice, affecting the olfactory pathway and limbic system, leading to hyposmia, anxiety, and memory loss.

The Membrane Interaction of Alpha-Synuclein

Cencen Liu et al.

Summary: The presynaptic protein alpha-synuclein (alpha-Syn), closely related to Parkinson's disease, has been extensively studied for its pathogenic mechanisms. Its interaction with membranes and influence on synaptic transmission may provide insights into both its pathology and physiological functions. Research on alpha-Syn and its variants interacting with membranes stands to expand current understandings of the protein.

Olfactory Signatures in the Food Finding Test in Mice With Normal and Alzheimer's Disease-Pathological Aging With Special Concerns on the Effects of Social Isolation

Daniela Marin-Pardo et al.

Summary: The temporal course and severity of sensory system involution through aging are critical for perceiving and recognizing the world. Olfactory loss is considered an early biomarker in Alzheimer's disease neurodegenerative process. In the 3xTg-AD model, olfactory patterns were found to differ between males and females, with social isolation impacting these patterns.

α-Synuclein aggregation in the olfactory bulb induces olfactory deficits by perturbing granule cells and granular-mitral synaptic transmission

Fengjiao Chen et al.

Summary: The aggregation of alpha-synuclein in the olfactory bulb can impair odor detection and perception in mice, as well as increase the activity of M/Ts cells. This study provides direct evidence of the impact of alpha-synuclein aggregates on olfactory dysfunction in Parkinson's disease and reveals the neural circuit mechanisms involved.

An Evolutionary Microcircuit Approach to the Neural Basis of High Dimensional Sensory Processing in Olfaction

Gordon M. Shepherd et al.

Summary: The processing of odor stimuli in mammals involves only two regions, the olfactory bulb and olfactory cortex, suggesting a highly efficient system for odor perception. Evolutionary insights from paleontology indicate that the basic architecture of the olfactory pathway has remained consistent for hundreds of millions of years, driven by olfactory receptor gene duplications. Multiple microcircuits with over 20 sequences are identified in the high dimensional processing of odor information.

Visualization of early oligomeric α-synuclein pathology and its impact on the dopaminergic system in the (Thy-1)-h[A30P]α-syn transgenic mouse model

Anish Behere et al.

Summary: The study revealed an abundance of previously undetected α-syn oligomers in the midbrain of young A30P transgenic mice, while phosphorylated and proteinase k-resistant α-syn species were observed to a larger extent in aged mice. Although no loss of dopaminergic neurons was detected in A30P transgenic mice, dysregulation in the monoaminergic system was recorded in older mice.

Overexpression of α-synuclein inhibits mitochondrial Ca2+ trafficking between the endoplasmic reticulum and mitochondria through MAMs by altering the GRP75-IP3R interaction

Adolfo Garcia Erustes et al.

Summary: The study showed that alpha-synuclein affects the MAMs and Ca2+ transfer from the ER to mitochondria, leading to mitochondrial membrane potential loss, increased mitophagy, and mitochondrial Ca2+ overload. Overexpression of alpha-synuclein also resulted in a reduction in ER-mitochondria contact sites and disrupted GRP75-IP3R interaction, indicating a dysregulation in MAM activity.
