Idebenone Regulates Aβ and LPS-Induced Neurogliosis and Cognitive Function Through Inhibition of NLRP3 Inflammasome/IL-1β Axis Activation

Hyun-ju Lee et al.

Summary: This study explores the effects of idebenone on neuroinflammatory responses, cognitive function, and underlying molecular mechanisms. The results suggest that idebenone can enhance neuroprotective NRF2 expression, reduce neuroinflammation, and improve learning and memory in mice. It also targets NLRP3 inflammasome activation and decreases gliosis and proinflammatory cytokine levels. In a mouse model of Alzheimer's disease, idebenone improves cognitive dysfunction and inhibits neuroinflammation.

Impairment of the autophagy-lysosomal pathway in Alzheimer's diseases: Pathogenic mechanisms and therapeutic potential

Wei Zhang et al.

Summary: Alzheimer's disease is a common neurodegenerative disorder characterized by memory loss and cognitive dysfunction. Impaired autophagy-lysosomal pathway is linked to AD pathogenesis, and enhancing autophagy to remove protein aggregates is proposed as a promising therapeutic strategy.

Faulty autolysosome acidification in Alzheimer's disease mouse models induces autophagic build-up of Aβ in neurons, yielding senile plaques

Ju-Hyun Lee et al.

Summary: This study reveals unique autophagy dysregulation within neurons in Alzheimer's disease mouse models, particularly deficient autolysosome acidification, leading to the accumulation of autophagic vacuoles and the formation of flower-like structures called "PANTHOS," which may contribute to the development of senile plaques in Alzheimer's disease.

Targeting microglial autophagic degradation of the NLRP3 inflammasome for identification of thonningianin A in Alzheimer's disease

Xiao-Gang Zhou et al.

Summary: The study identified thonningianin A as a potent microglial autophagy enhancer in Penthorum chinense Pursh, promoting the autophagic degradation of the NLRP3 inflammasome via the AMPK/ULK1 and Raf/MEK/ERK signaling pathways, providing novel insights for the treatment of Alzheimer's disease.

Transcription factor EB: an emerging drug target for neurodegenerative disorders

Ju-Xian Song et al.

Summary: TFEB as a master regulator of the autophagy-lysosomal pathway holds therapeutic potential for neurodegenerative disorders, with small molecules identified as TFEB activators showing promise in protecting against these diseases. Research is ongoing and the development of specific and potent TFEB activators with ideal brain bioavailability could provide a method for treating neurodegenerative disorders.

Targeting microglial autophagic degradation in NLRP3 inflammasome-mediated neurodegenerative diseases

An-Guo Wu et al.

Summary: Neuroinflammation is considered detrimental in neurodegenerative diseases, with NLRP3 inflammasome playing a crucial role in exacerbating pathology and disease progression. Microglial autophagy is important in maintaining brain homeostasis and negatively regulating NLRP3 inflammasome-mediated neuroinflammation. Understanding sex differences in NLRP3 inflammasome-mediated neurodegenerative diseases will aid in developing targeted therapies and improving diagnostic and prognostic abilities.

Electroacupuncture ameliorates beta-amyloid pathology and cognitive impairment in Alzheimer disease via a novel mechanism involving activation of TFEB (transcription factor EB)

Xiaoyan Zheng et al.

Summary: This study demonstrated the cognitive-enhancing effect of three-needle electroacupuncture in an animal model of Alzheimer's disease. The treatment reduced protein aggregation and inhibited glial cell activation by activating TFEB and promoting ALP in the brain. This suggests that three-needle electroacupuncture could be a promising therapy for Alzheimer's disease through a novel mechanism involving TFEB activation.

AMPK-dependent phosphorylation is required for transcriptional activation of TFEB and TFE3

Mathieu Paquette et al.

Summary: It has been shown that AMPK regulates the transcriptional activity of TFEB and TFE3 through phosphorylation, with a significant impact during nutrient deprivation, FLCN depletion, and pharmacological manipulation of MTORC1 or AMPK. MTORC1 specifically controls the cytosolic retention of TFEB and TFE3, while AMPK is essential for their transcriptional activity. This dual and opposing regulation mechanism is reminiscent of the regulation of ULK1, another key regulator of autophagy.
