Cholinergic and Neuroimmune Signaling Interact to Impact Adult Hippocampal Neurogenesis and Alcohol Pathology Across Development

Victoria A. Macht et al.

Summary: Alcohol use and misuse can have significant effects on individuals, especially when it begins during adolescence. It disrupts neurogenesis in the brain, and the severity of these effects depends on the timing of exposure and assessment. The cholinergic system, which regulates neurogenesis, is also affected by alcohol exposure. This review examines how alcohol exposure during critical developmental periods disrupts the maturation of the cholinergic and neuroinflammatory systems, leading to different effects on neurogenesis in adulthood.

Chronic alcohol drinking persistently suppresses thalamostriatal excitation of cholinergic neurons to impair cognitive flexibility

Tengfei Ma et al.

Summary: Exposure to chronic alcohol consumption impairs cognitive flexibility in individuals. This impairment is linked to altered activity of striatal cholinergic interneurons (CINs), which play a role in regulating glutamatergic transmission in medium spiny neurons (MSNs). Chronic alcohol drinking reduces thalamic excitation of CINs, compromising their ability to regulate glutamatergic transmission in MSNs, leading to impaired cognitive flexibility.

Association of Neural Reward Circuitry Function With Response to Psychotherapy in Youths With Anxiety Disorders

Stefanie L. Sequeira et al.

Summary: The study identified a relationship between neural activation to rewards and response to psychotherapy in youths with anxiety disorders, showing differences compared to healthy youths. Activation in the striatum to rewards did not differentiate between anxiety and healthy youths, but greater striatal responsivity to rewards may help youths with anxiety improve during treatment. This suggests that function in reward circuitry may guide the development of treatments for youths with anxiety.

Parkin Prevents Glutamate Excitotoxicity Through Inhibiting NLRP3 Inflammasome in Retinal Ganglion Cells

Xiuquan Wu et al.

Summary: This study found that upregulating Parkin attenuated glutamate-induced damage to retinal ganglion cells by inhibiting the NLRP3 inflammasome, while genetic and pharmacological downregulation of NLRP3 improved RGCs survival against glutamate excitotoxicity. Knockdown of Parkin exacerbated RGCs damage induced by glutamate, highlighting promising molecular targets for the prevention and treatment of acute optic nerve injury.

Crosstalk Among NLRP3 Inflammasome, ETBR Signaling, and miRNAs in Stress-Induced Depression-Like Behavior: a Modulatory Role for SGLT2 Inhibitors

Radwa N. Muhammad et al.

Summary: Depression is a major health concern with limited treatment efficacy, and the study suggests that dapagliflozin may have antidepressant potential through modulation of the NLRP3/ET-1/ETBR/BDNF/ZO-1 axis, particularly through ETBR signaling, to enhance BBB integrity, promote neuroplasticity, and exert antidepressant effects.

Ketogenic diet reduces alcohol withdrawal symptoms in humans and alcohol intake in rodents

Corinde E. Wiers et al.

Summary: This study suggests that a ketogenic diet may help in managing alcohol withdrawal and reducing alcohol consumption by decreasing the need for benzodiazepines during withdrawal, lowering cravings for alcohol, and improving brain bioenergetics.

nNOS-mediated protein-protein interactions: promising targets for treating neurological and neuropsychiatric disorders

Summary: Inhibiting nNOS can benefit various neurological and neuropsychiatric disorders, but direct inhibition may lead to severe side effects. Therefore, intervening in nNOS-mediated protein-protein interactions may be a more preferable option.
