Understanding these pathways has led to universal principles for targeted identification of peptide biosynthetic blueprints and rational engineering of biosynthetic pathways. This review contrasts the key principles of both routes and compares different strategies for peptide modifications.

Biosynthetic Elucidation and Structural Revision of Brevione E: Characterization of the Key Dioxygenase for Pathway Branching from Setosusin Biosynthesis

Summary: In this study, the biosynthetic gene cluster of fungal hexacyclic meroditerpenoid Brevione E (1) was identified, and its biosynthetic pathway was elucidated. The structure of 1 was found to have a substituted tetrahydrofuran ring instead of the previously proposed oxepane system. The key enzyme BrvJ, which is highly homologous to SetK, was determined to catalyze the skeletal rearrangement involved in cycloheptenone synthesis. This research provides insights into the divergence of biosynthetic pathways and natural product diversification in fungi.

Discovery of branching meroterpenoid biosynthetic pathways in Aspergillus insuetus: involvement of two terpene cyclases with distinct cyclization modes

Summary: A new meroterpenoid biosynthetic gene cluster was discovered in Aspergillus insuetus CBS 107.25, where a terpene cyclase folds the precursor compound 1 into a new meroterpenoid species with a different conformation. Another cyclase in the strain also synthesizes a distinct cyclized product of 1, resulting in diverging of the biosynthetic pathway in the fungus.

Molecular and Computational Bases for Spirofuranone Formation in Setosusin Biosynthesis

Summary: This study identified the biosynthetic gene cluster of Setosusin in the fungus Aspergillus duricaulis and elucidated its biosynthetic pathway through enzyme reconstitution experiments. The crucial role of the cytochrome P450 enzyme SetF in spirofuranone synthesis was revealed, showing its involvement in epoxidation and structural rearrangement processes. Mutagenesis experiments identified Lys303 as a potential catalytic residue important for spirofuranone synthesis.
