Biosynthesis of Guatrypmethine C Implies Two Different Oxidases for exo Double Bond Installation at the Diketopiperazine Ring

Summary: The study involved heterologous expression of a silent gene cluster from Streptomyces cinnamoneus which led to the identification of guatrypmethine C, a guaninylated cyclo-L-Trp-L-Met derivative with two exo double bonds at the diketopiperazine ring. Structural elucidation was achieved using MS, H-1, C-13, and N-15 NMR analyses including H-1-C-13 and H-1-N-15 HMBC. Gene combination and biochemical investigation confirmed that the formation of the two C=C bonds are catalyzed by two distinct enzyme families (i.e., cyclodipeptide oxidase and Fe-II/2OG-dependent oxygenase).

Global Genome Mining Reveals a Cytochrome P450-Catalyzed Cyclization of Crownlike Cyclodipeptides with Neuroprotective Activity

Summary: By conducting global genome mining of bacterial genomes, 829 cyclodipeptide biosynthesis gene clusters containing a cytochrome P450 gene were identified. Heterologous expression of these gene clusters led to the discovery of two novel crownlike cyclodipeptides, cyctetryptomycin A and B, which exhibit complex macrocycle structures and neuroprotective activity. Two cytochrome P450 enzymes catalyze sequential reactions resulting in the cyclization of diketopiperazine dimers within the gene clusters.
