Structural-based virtual screening and identification of novel potent antimicrobial compounds against YsxC of Staphylococcus aureus

The findings provide valuable insights for designing novel compounds against S. aureus.

Discovery of novel 4-phenylquinazoline-based BRD4 inhibitors for cardiac fibrosis

Summary: Bromodomain containing protein 4 (BRD4) is an epigenetic reader that has emerged as a potential therapeutic target for various diseases. Through high-throughput virtual screening and structure-activity relationship studies, a series of novel compounds targeting BRD4 were developed. Among them, compound C-34 showed better pharmacokinetic and physicochemical properties and exhibited lower cytotoxicity in vitro and in vivo, confirming its potential as a therapeutic agent for fibrotic cardiovascular disease.

Discovery of selective HDAC/BRD4 dual inhibitors as epigenetic probes

Summary: Selective HDAC/BRD4 dual inhibitors were designed and synthesized using pharmacophore fusion strategy, leading to the identification of three types of inhibitors with different cellular activities. Among them, the selective dual inhibitor 41c showed synergistic effects in inducing cell cycle arrest and apoptosis in MV-4-11 cells. This study supports the use of selective HDAC/BRD4 dual inhibitors as epigenetic probes for future biological evaluation in various cancer cell lines.

This work introduces a semi-automated approach to reparametrize the CHARMM lipid force field with consistent inclusion of long-range dispersion through the Lennard-Jones particle-mesh Ewald method. With careful selection of target properties and reweighting settings, the newly optimized lipid force field, denoted as C36/LJ-PME, successfully incorporates the explicit treatment of long-range dispersion.
