Discovery of novel β-carboline derivatives as selective AChE inhibitors with GSK-3β inhibitory property for the treatment of Alzheimer's disease

Summary: This study identifies a novel series of harmine derivatives with potential value for the treatment of Alzheimer's disease (AD). Compound ZLWH-23 shows significant anti-acetylcholinesterase activity, selective inhibition of butyrylcholinesterase, and acceptable inhibition of glycogen synthase kinase-3 beta. Furthermore, ZLWH-23 exhibits good selectivity for GSK-3 beta over other kinases and reduces tau hyperphosphorylation in a cell model. Harmine-based derivatives could be considered as drug leads for AD therapies.

Synthesis, kinetic evaluation and molecular docking studies of donepezil-based acetylcholinesterase inhibitors

Summary: New therapeutic agents for treating Alzheimer's disease were developed by designing and synthesizing donepezil-based analogs, which showed equally potent inhibitory activity against electric eel AChE by replacing the phenyl moiety with a pyridine ring. The most potent inhibitor demonstrated a dual-binding mode similar to donepezil, and molecular modeling studies suggested involvement of additional residues in the human AChE enzyme active site compared to donepezil.

Anticholinesterase activity of β-carboline-1,3,5-triazine hybrids

Summary: The activity of new compounds against acetylcholinesterase and butyrylcholinesterase was evaluated. The results showed that these compounds exhibited selectivity towards butyrylcholinesterase and demonstrated strong inhibitory activity. Among them, compound 12 showed the highest inhibitory activity and competed with butyrylthiocholine for the same binding site.

Design, synthesis and biological evaluation of harmine derivatives as potent GSK-3β/DYRK1A dual inhibitors for the treatment of Alzheimer's disease

Summary: Harmine derivatives were designed, synthesized, and evaluated as dual GSK-3 beta/DYRK1A inhibitors, showing promising activity against both targets. Compound ZDWX-25 exhibited potent inhibitory effects on GSK-3 beta and DYRK1A, with IC50 values of 71 and 103 nM, respectively. These compounds have the potential to be promising candidates for Alzheimer's disease treatment.
