Design, synthesis, and in silico studies of quinoline-based-benzo[d]imidazole bearing different acetamide derivatives as potent α-glucosidase inhibitors

Summary: In this study, 18 novel quinoline-based-benzo[d]imidazole derivatives were synthesized and screened for their alpha-glucosidase inhibitory potential. Except for 9q, all compounds in the series exhibited significant α-glucosidase inhibition, with compound 9d showing the most potent competitive inhibition. Molecular docking study indicated effective binding interactions between the designed compounds, including 9d, and the active site of the alpha-glucosidase enzyme.

Summary: The benzamide-based structural analogues showed potent inhibition of alpha-glucosidase, with most compounds being non-cytotoxic and demonstrating significant DPPH radical scavenging activity. Compound 9 exhibited cytotoxicity against 3T3 normal mouse fibroblast cell lines. Mode of enzyme inhibition was determined for some compounds based on kinetic studies.

Celebrex derivatives: Synthesis, α-glucosidase inhibition, crystal structures and molecular docking studies

Summary: Celebrex, commonly used as an anti-inflammatory drug, was functionalized to identify new alpha-glucosidase inhibitors. Many synthesized derivatives showed potent alpha-glucosidase inhibitory activity in vitro, with compound 9 being the most effective inhibitor. Molecular docking studies revealed interactions of compounds 6 and 9 with key amino acid residues of alpha-glucosidase.
