Perturbing HIV-1 Ribosomal Frameshifting Frequency Reveals a cis Preference for Gag-Pol Incorporation into Assembling Virions

Summary: This study reveals the mechanisms of Gag-Pol protein incorporation into HIV-1 virions. The study found that GP is enriched in virions about 3-fold compared to cells, and viral infectivity is better maintained at subphysiological levels of GP. It was also discovered that GP is more efficiently incorporated into virions when synthesized from the same mRNA as Gag. Furthermore, virions exhibit a strong upper limit to trans-delivered GP incorporation. These results provide new insights into the importance and regulation of GP in HIV-1 virion assembly.

The results show that m(6)A deposition leads to increased Gag synthesis and reduced packaging, while FTO-mediated demethylation promotes the incorporation of the full-length RNA into viral particles.

Our findings provide insights into the late stages of HIV-1 life cycle and unravel the mechanistic link between RNA dimerization, Pr55Gag binding, and genome packaging.

Strain and rupture of HIV-1 capsids during uncoating

Summary: Viral replication in HIV-1 relies on a fullerene-shaped capsid to transport genetic material deep into the nucleus of an infected cell. Capsid stability is linked to the presence of cofactors, including inositol hexakisphosphates (IP6) that bind to pores found in the capsid. This study used molecular dynamics simulations to analyze the strain patterns on capsid lattices and found that the presence of cofactors increased the rigidity of the capsid. Further analysis revealed that the capsid undergoes lattice strain and eventually ruptures during the reverse transcription process.
