Escaping from Flatland: Stereoconvergent Synthesis of Three-Dimensional Scaffolds via Ruthenium(II)-Catalyzed Noyori-Ikariya Transfer Hydrogenation

Summary: Noyori-Ikariya-type ruthenium(II)-catalysts have been used for 25 years in asymmetric transfer hydrogenation for the synthesis of chiral secondary alcohols. Recent progress has been made in understanding the asymmetric reduction of complex ketones, controlling up to four stereocenters in a single transformation. Intriguing multi-chiral molecular architectures can be prepared via ATH for drug discovery.

New dual ATP-competitive inhibitors of bacterial DNA gyrase and topoisomerase IV active against ESKAPE pathogens

Summary: The rise in multidrug-resistant bacteria highlights the need for new antibacterial agents less prone to resistance. Compounds simultaneously inhibiting multiple bacterial targets, like 31h, show promising antibacterial activities without cytotoxicity and potency against various pathogens, including those resistant to other drugs. The structural derivatives of 31h could represent a step towards clinically efficacious multitargeting antimicrobials resilient to existing antimicrobial resistance.

Practical Synthesis and Application of Halogen-Doped Pyrrole Building Blocks

Summary: In this study, a practical method for obtaining four new halogen-substituted pyrrole building blocks was developed, with synthetic steps ranging from two to five. The target compounds were prepared in quantities ranging from 50 mg to 1 g, and three of them were shown to be effective nanomolar inhibitors of bacterial DNA gyrase B, highlighting the potential utility of these chemical motifs in medicinal chemistry.
