BET-HDAC Dual Inhibitors for Combinational Treatment of Breast Cancer and Concurrent Candidiasis

Heat shock protein 90 (Hsp90)/Histone deacetylase (HDAC) dual inhibitors for the treatment of azoles-resistant Candida albicans

Summary: This study demonstrates that dual inhibition of heat shock protein 90 and histone deacetylase potentiates the efficacy of fluconazole against resistant Candida albicans infections. The first generation of Hsp90/HDAC dual inhibitors show synergistic enhancement to treat azoles-resistant candidiasis. These dual inhibitors represent a new strategy for the development of novel antifungal therapeutics to combat azole-resistant candidiasis.

Novel Carboline Fungal Histone Deacetylase (HDAC) Inhibitors for Combinational Treatment of Azole-Resistant Candidiasis

Summary: A series of novel carboline fungal HDAC inhibitors were designed and synthesized to combat antifungal drug resistance, with compound D12 showing potent synergistic effects with fluconazole against resistant Candida albicans infection. D12 reduced the virulence of C. albicans by blocking morphological mutual transformation and inhibiting biofilm formation, and reversed drug resistance by downregulating the expression of the azole target gene ERG11 and efflux gene CDR1.

This highlights the potential of CYP51/HDAC dual inhibitors as a promising strategy for the discovery of novel broad-spectrum antifungal agents.
