Design, synthesis and evaluation of 3-amide-5-aryl benzoic acid derivatives as novel P2Y14R antagonists with potential high efficiency against acute gouty arthritis

Ran Lu et al.

Summary: The P2Y(14) nucleotide receptor plays important roles in physiological and pathologic events, particularly in immune and inflammation. A series of novel P2Y(14) antagonists were designed based on the 3-amide benzoic acid scaffold, with compound 11m showing the most potent activity and improved pharmacokinetic properties. Compound 11m has promising efficacy in alleviating acute gouty arthritis and may be a potential therapeutic agent for intervention.

P2Y14 Receptor as a Target for Neutrophilia Attenuation in Severe COVID-19 Cases: From Hematopoietic Stem Cell Recruitment and Chemotaxis to Thrombo-inflammation

Lyvia Lintzmaier Petiz et al.

Summary: The global SARS-CoV-2 pandemic has resulted in over 2.3 million deaths, and effective treatment for COVID-19 is still lacking. The differentiation of hematopoietic stem cells is affected by COVID-19, leading to immune cell activation, inflammation, and excessive release of reactive oxygen species, influencing neutrophil activity.

Immunopathogenesis and treatment of cytokine storm in COVID-19

Jae Seok Kim et al.

Summary: Severe COVID-19 is characterized by systemic hyper-inflammation and cytokine storm, often leading to acute respiratory distress syndrome and multiple organ failure. Treatment of COVID-19 cytokine storm is crucial, with anti-inflammatory therapies playing a significant role. Various approaches are being used, including new cytokine-targeted therapies, to address the cytokine storm associated with COVID-19.
