Tumor-Cell-Specific Targeting of Ibrutinib: Introducing Electrostatic Antibody-Inhibitor Conjugates (AiCs)

Andreas Faust et al.

Summary: Linking ibrutinib with a cell-targeted internalizing antibody can reduce its toxicities and enhance pharmacological effectiveness. By complexing ibrutinib with an anti-CD20-protamine targeting conjugate and free protamine, a spontaneously assembling electrostatically stabilized vesicular nanocarrier is formed, leading to increased accumulation of the drug in target cells and amplified pharmacological effectiveness.

Targeted siRNA nanocarrier: a platform technology for cancer treatment

Nicole Baeumer et al.

Summary: A study found that the small arginine-rich protein protamine can condense genomic DNA into the sperm head, and its high RNA binding capacity can be utilized for the spontaneous electrostatic assembly of therapeutic nanoparticles decorated with tumor-cell-specific antibodies for efficient targeting of siRNA. These nanoparticles showed promising results in treating non-small-cell lung cancer and Ewing sarcoma models.

Identifying of biomarkers associated with gastric cancer based on 11 topological analysis methods of CytoHubba

Hua Ma et al.

Summary: This study identified 11 hub genes associated with gastric cancer, with functions enriched in cellular senescence, cell cycle, viral carcinogenesis, and the p53 signaling pathway. Ten of these genes were found to be related to overall survival in GC patients, and seven were significantly correlated with GC, potentially serving as prognostic biomarkers.

Summary: This study identified 14 hub genes in HCC whose expression levels were significantly up-regulated and negatively correlated with overall survival time, and were also positively correlated with immune infiltration cells. Among these hub genes, KIF2C and UBE2C showed the most significant correlation with immune cell infiltration in HCC, suggesting their potential as prognostic biomarkers for guiding immunotherapy.

MDM2 inhibitor APG-115 exerts potent antitumor activity and synergizes with standard-of-care agents in preclinical acute myeloid leukemia models

Douglas D. Fang et al.

Summary: Acute myeloid leukemia (AML) is a clinically and genetically heterogeneous clonal disease with unmet medical needs. Targeting the MDM2-P53 axis to restore P53 function has emerged as an attractive AML treatment strategy. APG-115, a potent MDM2 inhibitor, has demonstrated substantial antileukemic activity in AML cellular cultures and animal models.

Hotspot DNMT3A mutations in clonal hematopoiesis and acute myeloid leukemia sensitize cells to azacytidine via viral mimicry response

Marina Scheller et al.

Summary: The study showed that cancer cells with hotspot DNMT3A mutations are sensitive to the DNMT1 inhibitor azacitidine through focal DNA demethylation, viral mimicry, and interferon activation. This suggests that DNMT3A(R882) mutations could potentially be a predictive marker for azacitidine response in acute myeloid leukemia.
