Novel piperine-carboximidamide hybrids: design, synthesis, and antiproliferative activity via a multi-targeted inhibitory pathway

Discovery of new pyrimido[5,4-c]quinolines as potential antiproliferative agents with multitarget actions: Rapid synthesis, docking, and ADME studies

Summary: A series of novel pyrimido[5,4-c]quinoline derivatives were synthesized and demonstrated to have in vitro antiproliferative activity. The compounds were evaluated for their inhibition of topoisomerase I, topoisomerase II alpha, CDK2, and EGFR. The results showed selective inhibitory activity against topoisomerase I and weak inhibitory activity against topoisomerase II alpha. Compound 30B exhibited the most potent anti-proliferative activity and moderate inhibition of CDK2. Compounds 30B, 28d, and 28c also showed significant inhibitory activity against EGFR.

Novel indazole derivatives as potent apoptotic antiproliferative agents by multi-targeted mechanism: Synthesis and biological evaluation

Summary: Indazole is an important class of heterocyclic compounds with diverse biological activity. In this study, a novel series of differently substituted indazole derivatives 6a-v were synthesized and evaluated for their biological activity. The results demonstrated that compounds 6f, 6i, 6j, 6s, and 6n exhibited significant antiproliferative activity against cancer cells and showed promising potential as potent antiproliferative agents by inhibiting EGFR, CDK2, and c-Met and inducing apoptosis through cytochrome C overexpression and activation of the intrinsic apoptotic pathway.

Summary: A series of new quinoline/chalcone hybrids containing 1,2,4-triazole moiety were designed and synthesized in this study, showing moderate to good activity against various cancer cell lines. Some of these compounds exhibited promising antiproliferative activities and displayed high binding affinities in inhibiting EGFR and BRAF(V600E) kinases.

Summary: A series of novel piperine-resveratrol hybrids 5a-h were designed, synthesized, and evaluated for their antiproliferative activities against cancer cell lines. Compound 5b, possessing resveratrol pharmacophoric phenolic moieties, demonstrated strong inhibition against leukemia and breast cancer cell lines, with the strongest inhibition of Sirt-2 recorded. The study provides insights into the potential therapeutic implications of these hybrids as selective Sirt-2 inhibitors.
