A review on synthetic chalcone derivatives as tubulin polymerisation inhibitors

Wenjing Liu et al.

Summary: Chalcone derivatives have been found to disrupt the dynamic balance of microtubules, inhibit tumor cell proliferation, and exhibit anti-tumor effects. By inhibiting microtubule dynamics and disrupting microtubule homeostasis, they interfere with the process of cell mitosis.

In vivo- and in silico-driven identification of novel synthetic quinoxalines as anticonvulsants and AMPA inhibitors

Hamada S. Abulkhair et al.

Summary: This study reported the design and synthesis of two novel sets of quinoxaline derivatives with potential anticonvulsant activity, showing that some of the synthesized compounds exhibited promising activities. Docking studies suggested that the mechanism of action of these derivatives could be AMPA binding.

Discovery of novel indole derivatives that inhibit NEDDylation and MAPK pathways against gastric cancer MGC803 cells

Dong-Jun Fu et al.

Summary: The novel indole derivative V7 showed potent inhibitory activity against MGC803 cancer cells by inhibiting colony formation, inducing apoptosis, and arresting cell cycle at G2/M phase. Moreover, V7 also inhibited the NEDDy-lation pathway and MAPK pathway against MGC803 cells.

Reactive oxygen species (ROS)-response nanomedicine through knocking down a novel therapeutic target NEDD8-conjugating enzyme UBC12 (UBE2M) in the treatment of liver cancer

Zhanxia Zhang et al.

Summary: This study developed a folic acid-modified PLGA-TK-PEG nanomedicine for siRNA delivery, achieving targeted therapy for liver cancer cells. By releasing siRNA in the tumor microenvironment, the nanomedicine not only inhibits the Neddylation pathway but also promotes the accumulation of tumor-suppressive substrates.

Recent Progress on Tubulin Inhibitors with Dual Targeting Capabilities for Cancer Therapy

Wen Shuai et al.

Summary: Microtubules are crucial in cellular functions and important targets for cancer therapy, but drug resistance and toxicity limit their efficacy. Dual-target drugs, combining tubulin with other anti-tumor agents, are considered promising to overcome limitations and improve therapeutic efficacy. Rational target combinations and design strategies of dual-target tubulin inhibitors are discussed for future directions.
