A framework for standardized qPCR-targets and protocols for quantifying antibiotic resistance in surface water, recycled water and wastewater

Ishi Keenum et al.

Summary: Water environments are recognized as a conduit for the spread of antibiotic resistance, and there is a need to standardize monitoring protocols. qPCR is a sensitive method for quantifying antibiotic resistance genes, but quality assurance and other aspects of protocols need to be addressed. Identified gene targets in water samples include sul1, tetA, and intI1, with vanA and blaCTX-M as clinically-relevant targets.

Fate and seasonality of antimicrobial resistance genes during full-scale anaerobic digestion of cattle manure across seven livestock production facilities

Tucker R. Burch et al.

Summary: This study investigated the fate of antibiotic resistance genes (ARGs) during anaerobic digestion of livestock manure. The results showed that concentrations of intI1, sul1, and tet(A) decreased during anaerobic digestion, but their removal was less extensive than expected. Furthermore, tet(W) concentrations remained unchanged and erm(B) concentrations increased. These findings suggest that mesophilic anaerobic digestion may have limitations as an intervention for ARGs in livestock manure.

Occurrence of Indicator Genes of Antimicrobial Resistance Contamination in the English Channel and North Sea Sectors and Interactions With Environmental Variables

Erwan Bourdonnais et al.

Summary: This study investigated the state of antimicrobial resistance and the abundance and distribution of resistance genes in the English Channel and North Sea areas. The results showed that the sul1 and intI1 genes were positively correlated with dissolved oxygen and turbidity, while the microbial population was correlated with pH, temperature, salinity, dissolved oxygen and turbidity. Furthermore, a sample from the West Netherlands coast area had high abundances of the tetA, sul1, and intI1 genes. This study is the first to demonstrate the impact of anthropogenic inputs and environmental variables on antimicrobial resistance contamination indicators in seawaters of the North Sea and English Channel.

Impacts of farmland application of antibiotic-contaminated manures on the occurrence of antibiotic residues and antibiotic resistance genes in soil: A meta-analysis study

Yu Zhang et al.

Summary: A meta-analysis was conducted to investigate the impacts of farmland application of antibiotic-contaminated manures on antibiotic concentrations and ARG abundances in manure-amended soil. The results showed that the use of antibiotic-contaminated manures significantly increased the levels of antibiotics and ARGs in the soil. Additionally, soil organic carbon and silt content were found to be correlated with antibiotic concentrations and ARG abundances.

Geospatial Patterns of Antimicrobial Resistance Genes in the US EPA National Rivers and Streams Assessment Survey

Scott P. Keely et al.

Summary: Antimicrobial resistance is a global problem caused by the excessive use of antimicrobials. The spread of antimicrobial resistance genes (ARGs) through the environment and their impact on microbial pathogens are not well understood. This study reveals that water bodies receiving waste discharge have higher risks of microbial exposure compared to less disturbed aquatic environments. The findings provide valuable information for assessing the effect of environmental ARGs on public health and implementing mitigation strategies.

Non-point source fecal contamination from aging wastewater infrastructure is a primary driver of antibiotic resistance in surface waters

Julian Damashek et al.

Summary: Antibiotic resistance poses a global threat to human health. This study examined the presence of fecal marker genes and antibiotic resistant genes (ARGs) in stream water samples from the Upper Oconee watershed in Georgia, USA. The results showed widespread fecal contamination from humans, ruminants, and poultry, with a high percentage of samples testing positive for at least one of the six targeted ARGs. Non-point sources of fecal contamination, particularly aging wastewater infrastructure, were identified as critical disseminators of ARGs in the environment.

Targeting current and future threats: recent methodological trends in environmental antimicrobial resistance research and their relationships to risk assessment

Tucker R. Burch et al.

Summary: Antimicrobial resistance (AMR) is a growing public health threat. Current highly-cited papers studying AMR in environmental engineering and agricultural systems primarily employ targeted methods, with a lack of studies using non-targeted methods. Improving AMR surveillance in environmental reservoirs requires risk assessment, and different monitoring approaches imply different objectives for risk assessment.

Treated wastewater irrigation promotes the spread of antibiotic resistance into subsoil pore-water

Ioannis D. Kampouris et al.

Summary: This study investigated the impact of treated wastewater (TWW) irrigation on antibiotic resistance genes (ARGs) in subsoil pore-water, finding that TWW irrigation increases ARG prevalence. The hypothesis was validated through field and laboratory experiments, showing that TWW irrigation promoted the spread of ARGs in subsoil pore-water.

Quantitative Microbial Risk Assessment for Contaminated Private Wells in the Fractured Dolomite Aquifer of Kewaunee County, Wisconsin

Tucker R. Burch et al.

Summary: This study investigated the health burden associated with contamination of private wells in Kewaunee County by feces-borne gastrointestinal pathogens. The results suggest that private well contamination could be responsible for as many as 301 acute gastrointestinal illness cases per year in the county, with bovine and human fecal sources being the main contributors. Cryptosporidium parvum was predicted to be the most common pathogen causing illness in the quantitative microbial risk assessment.

Sources and Risk Factors for Nitrate and Microbial Contamination of Private Household Wells in the Fractured Dolomite Aquifer of Northeastern Wisconsin

Mark A. Borchardt et al.

Summary: The study found that total coliforms and nitrate in groundwater were mainly associated with depth-to-bedrock at well sites and nearby agricultural land, not septic systems. Both human wastewater and cattle manure were contributors to well contamination. Nitrate and Escherichia coli were the most frequently detected contaminants.

The Environmental Microbiology Minimum Information (EMMI) Guidelines: qPCR and dPCR Quality and Reporting for Environmental Microbiology

Mark A. Borchardt et al.

Summary: Real-time quantitative polymerase chain reaction (qPCR) and digital PCR (dPCR) methods have revolutionized environmental microbiology by providing quantitative organism-specific data. However, many researchers in the field fail to report necessary controls and methods, impacting the reliability of their study results. By implementing systematic controls, efficient workflow, and critical reporting elements, data credibility in environmental microbiology research can be enhanced to support disease prevention efforts.

Antibiotic resistance genes in surface water and groundwater from mining affected environments

Hai-Yan Zou et al.

Summary: Mining activities are known to generate a large amount of mine tailings and mine drainage containing various heavy metals, which can lead to increased bacterial antibiotic resistance. This study found higher abundances of antibiotic resistance genes in mining-affected water, especially sulfonamides, chloramphenicols, and tetracyclines resistance genes, with positive correlations between heavy metals and ARGs. Additionally, the microbial community composition differed between mining-affected water and reference water, with certain dominant phyla being identified as potential hosts for the ARGs.

Unraveling the riverine antibiotic resistome: The downstream fate of anthropogenic inputs

Jangwoo Lee et al.

Summary: This study tracked the fate of the resistome in treated wastewater in two Swiss rivers using a comprehensive analysis and found that wastewater-origin ARG did not persist in rivers after 5-6.8 km downstream. The potential for direct human exposure to the resistome of wastewater-origin appeared to typically abate rapidly in the studied rivers.
