Macrophages transfer mitochondria to sensory neurons to resolve inflammatory pain

Michiel van der Vlist et al.

Summary: The resolution of inflammatory pain is actively controlled by macrophages through the transfer of mitochondria to sensory neurons. This process requires the expression of CD200R on macrophages and the non-canonical CD200R ligand iSec1 on sensory neurons.

Extracellular Vesicles Derived from Neural Progenitor Cells--a Preclinical Evaluation for Stroke Treatment in Mice

X. Zheng et al.

Summary: Stem cells such as mesenchymal stem cells (MSCs) enhance neurological recovery in preclinical stroke models by secreting extracellular vesicles (EVs), with neural progenitor cells (NPCs)-derived EVs showing therapeutic potential in reducing cell injury and promoting neuroregeneration in mice post-stroke. NPC-EVs delivery leads to increased blood concentrations of B and T lymphocytes without affecting cerebral cell counts, suggesting a potential contribution to reverting post-stroke immunosuppression. Furthermore, biodistribution analysis reveals the majority of NPC-EVs to be found in extracranial organs like the liver and lung after systemic administration.

Mesenchymal stromal cell extracellular vesicles rescue mitochondrial dysfunction and improve barrier integrity in clinically relevant models of ARDS

Johnatas Dutra Silva et al.

Summary: In acute respiratory distress syndrome (ARDS), mitochondrial dysfunction contributes to damage of the alveolar epithelial and endothelial barriers. Mesenchymal stromal cells-derived extracellular vesicles (MSC-EVs) can improve alveolar-capillary barrier integrity through mitochondrial transfer, restoring normal barrier properties and mitochondrial functions in a model of ARDS.

Microvesicles transfer mitochondria and increase mitochondrial function in brain endothelial cells

Anisha D'Souza et al.

Summary: It was shown for the first time that microvesicles derived from human brain endothelial cells can transfer polarized mitochondria to recipient cells and increase ATP levels. The study also revealed a high association to glycolysis-related processes and demonstrated the selective transfer of mitochondria to increase endothelial cell survival under ischemic conditions.

Heat shock protein 27 activity is linked to endothelial barrier recovery after proinflammatory GPCR-induced disruption

Cara C. Rada et al.

Summary: Vascular inflammation can disrupt endothelial barrier integrity and cause tissue edema through the activation of PAR1 and p38 MAPK signaling pathways. The phosphorylation and function of HSP27, a key protein regulating endothelial barrier integrity, are differentially controlled by effector kinases MK2 and MK3.

Exogenous inter-α inhibitor proteins prevent cell death and improve ischemic stroke outcomes in mice

Louise D. McCullough et al.

Summary: IAIPs have shown significant therapeutic effects in ischemic stroke, especially in delayed treatment, with potential benefits in different age groups of experimental animals. The therapeutic effects of IAIPs in ischemic stroke are mediated through mechanisms such as inhibiting C5aR1. These results suggest that IAIPs are a potential candidate for the treatment of ischemic stroke.
