Acid-Degradable Hydrogen-Generating Metal-Organic Framework for Overcoming Cancer Resistance/Metastasis and Off-Target Side Effects

Summary: The development of stimuli-responsively degradable porous carriers for controlled drug release is important for clinical translation. In this study, a new type of Fe-MOF nanocrystals is engineered as an acid-degradable drug carrier and hydrogen donor. Fe-MOF nanocrystals exhibit acid-responsive degradation for H-2 generation and simultaneous drug release, and can effectively inhibit cancer metastasis and sensitize MDR cancer cells for enhanced chemotherapy. The high anti-MDR/antimetastasis efficacies and biocompatibility make Fe-MOF nanocrystals and Fe-MOF-based nanomedicine highly promising for clinical translation.

Drug-Sponge Lipid Nanocarrier for in Situ Cargo Loading and Release Using Dynamic Covalent Chemistry

Summary: This study introduces a drug delivery strategy that involves reversible pH-controlled capture and release of active cargo based on dynamic covalent chemistry within lipid nano-droplets, termed as drug sponge. This approach effectively encapsulates fluorescent dyes and doxorubicin drugs, allowing for stable formulations at neutral pH and controlled release in acidic environments. The drug-sponge nanoemulsions exhibit cytotoxicity in cancer cells and inhibit tumor growth in mouse xenograft models, as well as enable unprecedented extraction of dye/drug cargos directly from cells and tissues.
