Enhancing the Antifungal Activity and Ophthalmic Transport of Fluconazole from PEGylated Polycaprolactone Loaded Nanoparticles

Summary: This study investigated the antifungal activity, ocular irritation, and transport of fluconazole-loaded poly (epsilon-caprolactone) nanoparticles in a rabbit eye model. The optimized nanoparticles showed excellent antifungal activity, high drug entrapment efficiency, and were able to be successfully delivered to the eye using a thermosensitive in situ hydrogel, without causing irritation to the cornea.

Summary: Pterygium is a progressive disease of the human eye with limited treatment options. Previous research suggests that curcumin, a compound derived from turmeric, shows promise in treating pterygium. However, curcumin has low solubility, which limits its effectiveness. A new nanoformulation called Cur-AgNPs, made by stabilizing curcumin with silver nanoparticles, offers a solution by increasing curcumin’s bioavailability without inducing toxicity. This new compound has the potential to deliver both curcumin and silver to the affected tissue simultaneously, presenting a more sophisticated strategy for the treatment of human pterygium.

This study evaluated different polyaphron formulations and found that the 3% polyquaternium 10-based formulation had the best corneal retention, while the 10% poloxamer 188 and 1% polyquaternium 10-based formulations were the most stable. Toxicological evaluations showed that all four formulations had good biocompatibility with ocular tissues. These drug delivery systems have great potential for ocular drug delivery.
