A guide of column selection for two-dimensional liquid chromatography method development of natural alkaloids

Summary: Natural products, especially alkaloids, are valuable drug leads in drug discovery. Two-dimensional liquid chromatography (2D-LC) is an efficient tool for analyzing and separating alkaloids. The diversity of alkaloids often leads to peak tailing issues in their separation. To address this problem, a guide for column selection and combination was proposed in this study, which helped improve alkaloid separation. The guide included a comprehensive evaluation of column applicability and orthogonality, providing a useful starting point for column selection in 2D-LC method development. A 2D-LC method was developed using the guide, which successfully separated an alkaloid sample from U. rhynchophylla with symmetrical peaks and high orthogonality. This evaluation process can also facilitate a quick understanding of the performance of newly designed stationary phases.

Summary: The study found that different drying methods significantly influence the chemical components in Panax notoginseng leaves (PNL), with sun-air drying and hot-air drying leading to vigorous ginsenoside transformation. Specifically, the main components of PNL can be transformed into different compounds by selective cleavage of beta-1,2-glucosidic linkage, with only steaming prior to drying able to inhibit this transformation. These findings provide a scientific basis for designing industrial drying methods to ensure PNL quality.

Chemical comparison of Astragali Radix by UHPLC/Q-TOF-MS with different growing patterns

Summary: This study compared cultivated Astragali Radix (AR-C) and wild/semi-wild Astragali Radix (AR-W) using ultra-high-performance liquid chromatography coupled with hybrid triple quadrupole time-of-flight mass spectrometry. Chemical investigation of Astragali Radix led to the tentative annotation of 196 compounds, and further analysis showed that 55 compounds had higher contents in the wild/semi-wild Astragali Radix. Thirteen compounds showed FC values higher than 3, and one of them could be used as a marker for discrimination of AR-C and AR-W. The study also found differences in the substitution of certain compounds between the two types of Astragali Radix.
