Synaptic density and cognitive performance in Alzheimer's disease: A PET imaging study with [11C]UCB-J

Summary: In this study, the SV2A abnormalities in patients with FCD II were evaluated using F-18-SynVesT-1 PET and compared with F-18-FDG PET. The results showed that F-18-SynVesT-1 PET had a higher accuracy for the localization of FCD II lesions than MRI and a more restricted pattern of abnormality than F-18-FDG PET.

Changes in synaptic density in the subacute phase after ischemic stroke: A 11C-UCB-J PET/MR study

Summary: This study used C-11-UCB-J PET imaging to discover a decrease in synaptic density in the ischemic tissue of stroke patients, as well as lower synaptic density in non-lesioned tissue of the affected hemisphere compared to the unaffected hemisphere. Furthermore, the contralesional cerebellar hemisphere also showed lower C-11-UCB-J SUVR compared to the ipsilesional cerebellar hemisphere, with some patients exhibiting crossed cerebellar diaschisis. Longitudinal research is needed to further study these changes in synaptic density and their relationship to outcomes.

Association of entorhinal cortical tau deposition and hippocampal synaptic density in older individuals with normal cognition and early Alzheimer's disease

Detection of changes in synaptic density in amyotrophic lateral sclerosis patients using 18F-SynVesT-1 positron emission tomography

Summary: This study aimed to evaluate the diagnostic value of F-18-SynVesT-1-PET as a marker of ALS pathology and investigate the differences in synaptic density between different ALS subtypes. The study found that ALS patients had lower synaptic density compared to healthy controls, and this was associated with cognitive impairment and site of onset. This approach may serve as a useful biomarker for ALS diagnosis, but currently it is not recommended as a routine investigation.

A 2-year study found that the accumulation of tau is associated with synaptic loss, and the regional distribution of synaptic loss aligns with the pattern of tau spread. Furthermore, changes in synaptic density are correlated with changes in cognitive function.
