SIRT1 selectively exerts the metabolic protective effects of hepatocyte nicotinamide phosphoribosyltransferase

Cassandra B. Higgins et al.

Summary: This study demonstrates the upregulation of hepatocyte NAMPT during fasting, which plays a role in regulating FGF21 activation and glucose homeostasis. Moreover, hepatic NAMPT can improve dyslipidemia and induce adipose browning in obese mice. Modulating hepatocyte NAD(+) may be a potential mechanism to attenuate fasting-responsive disease.

Structurally analogous trehalose and sucrose glycopolymers - comparative characterization and evaluation of their effects on insulin fibrillation

Malgorzata Milewska et al.

Summary: Trehalose glycopolymers have been extensively studied recently and have shown some unique properties. In order to compare their properties, researchers synthesized structurally similar glycopolymers composed of trehalose and sucrose. These glycopolymers have very low content of non-saccharide structural fragments, making their properties strongly influenced by the saccharide moieties. The study comprehensively characterized the glycopolymers, including their tendency to self-assemble, hydrolytic stabilities at different pH, and susceptibility to enzymatic hydrolysis.

Targeting hepatocyte carbohydrate transport to mimic fasting and calorie restriction

Summary: The pervasion of three daily meals and snacks has coincided with a rise in obesity and cardiometabolic disorders. Research shows that calorie restriction and intermittent fasting have beneficial effects on cardiometabolic and other diseases, but sustainable adherence to strict lifestyle changes is rare.

Impact of liver-specific GLUT8 silencing on fructose-induced inflammation and omega oxidation

Marta G. Novelle et al.

Summary: Excessive consumption of high-fructose diets can lead to insulin resistance, obesity, and NAFLD, with fructose affecting hepatic lipogenesis regulation differently in males and females. Further studies are needed to consider gender disparities in fructose-induced liver damage. GLUT8 is suggested as a potential target for preventing NAFLD, as its silencing has shown positive effects on liver inflammation and fatty acid content reduction in a fructose-induced non-alcoholic steatohepatitis female mouse model.

Trehalose causes low-grade lysosomal stress to activate TFEB and the autophagy-lysosome biogenesis response

Se-Jin Jeong et al.

Summary: The autophagy-lysosome system is crucial for recycling dysfunctional components in cells, and trehalose, a natural disaccharide, has the ability to induce autophagy by potentially affecting lysosomal function. This suggests that trehalose could serve as a therapeutic target for diseases associated with dysfunctional autophagy.

Summary: The study synthesized data on circulating TNF-alpha levels in patients with NAFLD and controls. It found that TNF-alpha levels were higher in NAFLD patients than controls, and were also associated with the severity of NAFLD. Additionally, male ratio was positively associated with TNF-alpha levels in the comparison between NASH and NAFL patients.

Summary: NAFLD is the most prevalent liver disease worldwide, with no approved pharmacological therapies available. The spectrum of severity ranges from steatosis to NASH, which is linked to increased risks of liver cancer and cirrhosis. Therefore, a holistic approach and ongoing therapeutic options are necessary to address the global health burden of these diseases.

Role of Tetraspanins in Hepatocellular Carcinoma

Sicheng Cai et al.

Summary: HCC is characterized by high prevalence, morbidity, and mortality, with tetraspanins playing important roles in its lifecycle. Some tetraspanins such as CD9, TSPAN15, and TSPAN31 can promote HCC cell proliferation or suppress apoptosis, while CD63, CD151, and TSPAN8 can facilitate HCC metastasis. TSPAN1, TSPAN8, and CD151 act as prognosis indicators and are inversely correlated to the overall survival rate of HCC patients.

Importance of Adipose Tissue NAD+ Biology in Regulating Metabolic Flexibility

Michael P. Franczyk et al.

Summary: The study showed that NAD(+) and NAMPT have a crucial role in regulating metabolic flexibility, with the loss of NAMPT leading to increased inflammation, decreased insulin sensitivity, and impaired glucose oxidation. This highlights the importance of NAD(+) biosynthesis in adipose tissue in maintaining metabolic health.
