Mechanistic insights into the nickel-dependent allosteric response of the Helicobacter pylori NikR transcription factor

Delineating the conformational landscape of the adenosine A2A receptor during G protein coupling

Summary: This study used fluorine nuclear magnetic resonance spectroscopy to identify key functional states of the adenosine A(2A) receptor complexed with heterotrimeric G protein in a phospholipid membrane environment. The findings revealed an ensemble consisting of inactive states, a G-protein-bound activation intermediate, and nucleotide-free states associated with agonist-driven activation. The crucial role of the G beta gamma subunit in facilitating ligand-dependent allosteric transmission was demonstrated.

Allosteric regulation of the nickel-responsive NikR transcription factor from Helicobacter pylori

Summary: Nickel plays a crucial role in the survival of the pathogenic bacteria Helicobacter pylori in the human stomach by regulating nickel homeostasis through the transcription factor NikR. Nickel binding induces conformational changes in HpNikR, enabling it to bind specific DNA motifs and influence gene transcription. Zinc is unable to induce the same allosteric response as nickel, suggesting that the mechanism of nickel-activated DNA binding by HpNikR is driven by conformational selection.
