Assessment of the genotoxic effects of antihypertensive drug active ingredient indapamide in human lymphocytes

Summary: This study aimed to assess the genotoxic effects of indapamide and found that it did not significantly affect chromosomal aberration and micronucleus frequencies, but caused significant damage in the comet assay at the highest concentration. It also increased sister chromatid exchange frequency at the two highest concentrations in the 48-h treatment. Overall, indapamide did not have significant genotoxic effects in these conditions.

Genotoxic effects of gadobutrol and gadoversetamide active substances used in magnetic resonance imaging in human peripheral lymphocytes in vitro

Summary: This study investigated the genotoxicity of gadobutrol and gadoversetamide, two gadolinium-based contrast agents commonly used in magnetic resonance imaging. The results showed that both agents are likely to pose a genotoxic risk in human peripheral lymphocytes, with gadoversetamide demonstrating slightly stronger effects than gadobutrol. Further research is needed to determine the specific concentrations and treatment periods for the safe usage of these contrast agents.

Assessment of genetic damage induced by gadolinium-based radiocontrast agents

Summary: This study aimed to investigate the genotoxic and cytotoxic potentials of gadoteric acid and gadoversetamide, two gadolinium-based contrast agents used in magnetic resonance imaging. The results showed that gadoteric acid did not have genotoxic or cytotoxic potential, while gadoversetamide might have both genotoxic and cytotoxic potential on human peripheral blood lymphocytes.

Gadolinium-Based Contrast Agents: Updates and Answers to Typical Questions Regarding Gadolinium Use

Summary: Gadolinium-based contrast agents have been widely used in magnetic resonance imaging to enhance diagnostic capabilities. While they have a good safety profile, there have been reported cases of nephrogenic systemic fibrosis in patients with advanced kidney disease. Recently, trace amounts of gadolinium deposition in the brain and other organs have also been found after contrast exposure, even in patients with normal renal function. This review provides an overview of recent updates and discusses typical clinical situations related to the use of gadolinium-based contrast agents.

Summary: Several studies over the past five years have reported deposition and retention of gadolinium in the brain and other organs after administration of gadolinium-based contrast agents during radiological procedures. However, the association between gadolinium accumulation and physical or neurological side effects has not been definitively demonstrated.

Biomarkers of geno- and cytotoxicity in the native broad-snouted caiman (Caiman latirostris): Chromosomal aberrations and mitotic index

Summary: The sensitivity of chromosomal aberration and mitotic index assays on Caiman latirostris peripheral blood lymphocytes following ex vivo exposure to the alkylating agent, MMS, was evaluated. The experiments showed that MMS exposure reduced the number of metaphases observed but increased the numbers of cells with MN and the percentages of aberrant metaphases. The incorporation of MI and CA tests in C. latirostris can provide valuable information on damage caused by xenobiotic exposures.

Comet assay: a versatile but complex tool in genotoxicity testing

Summary: The comet assay is a versatile method for measuring DNA strand breaks in individual cells, with modified versions able to detect oxidized DNA bases and reveal DNA damage induced indirectly by reactive oxygen species. Despite being semi-quantitative, there are possible approaches to improve DNA damage quantitation and enhance protocol standardization for comparability. The in vivo comet assay has an advantage over the micronucleus erythrocyte test as it can be applied to any organ and has been used in risk assessment for specific food contaminants.
