Successful treatment of post-traumatic stress disorder reverses DNA methylation marks

Christiaan H. Vinkers et al.

Summary: This study showed that successful treatment of PTSD is associated with significant changes in DNA methylation of certain genes, such as ZFP57. The longitudinal evidence suggests that ZFP57 methylation plays a role in both the development and successful treatment of deployment-related PTSD.

Placental DNA methylation marks are associated with maternal depressive symptoms during early pregnancy

Riikka J. Lund et al.

Summary: Maternal depressive symptoms during pregnancy can influence placental DNA methylation, particularly in genes related to neuronal development, which may impact the longterm health of the developing brain. The study found that maternal depressive symptoms are associated with DNA methylation marks in placenta, which are enriched for developmental genes and linked to mental disease traits. Further research is needed to determine if similar marks can be detected in exposed children.

DNA methylation in cord blood in association with prenatal depressive symptoms

Theodora Kunovac Kallak et al.

Summary: The study found differential DNA methylation in children exposed to comorbid prenatal depression and anxiety symptoms compared to healthy controls at some genes, while no significant differences were observed in children exposed to prenatal depression alone. Additionally, significant DNA methylation differences were found in children not receiving SSRI treatment and healthy controls compared to those exposed to SSRIs at certain gene sites and regions.

Summary: This study found that greater COVID-19-related prenatal stress was significantly associated with higher infants' SLC6A4 methylation in seven CpG sites, and SLC6A4 methylation at these sites predicted infants' temperament at 3 months.

Statistical methods for mediation analysis in the era of high-throughput genomics: Current successes and future challenges

Ping Zeng et al.

Summary: Mediation analysis investigates the intermediate mechanism by which an exposure influences the outcome. It is increasingly popular in high-throughput genomics studies, but faces challenges in genomics due to the large number of potential mediators and the composite null nature of the mediation effect hypothesis. New high-dimensional mediation methods have been developed to address these challenges, providing guidance for statisticians and computational biologists.
