Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Neoadjuvant Chemotherapy Induces IL34 Signaling and Promotes Chemoresistance via Tumor-Associated Macrophage Polarization in Esophageal Squamous Cell Carcinoma

Shotaro Nakajima et al.

Summary: The study revealed that NAC significantly increased the expression of CD163 on TAMs in ESCC, and IL34 played a crucial role in this effect. High expression of IL34 in ESCC patients was associated with poor prognosis, and IL34 expression was regulated by NAC, shifting the TME towards an immunosuppressive environment.

Interferon- and STING-independent induction of type I interferon stimulated genes during fractionated irradiation

Ruben S. A. Goedegebuure et al.

Summary: Clinically applied fractionated low-dose irradiation induces a delayed type I interferon response independently of interferon expression or STING signaling. These findings have implications for targeting the type I interferon response in cancer treatment. Time-course analyses showed a peak in interferon-stimulated gene expression within 2-3 weeks of treatment.

9p21 loss confers a cold tumor immune microenvironment and primary resistance to immune checkpoint therapy

Guangchun Han et al.

Summary: This study found that the loss of 9p21.3 leads to a cold tumor immune phenotype, reducing the responsiveness of tumors to immune checkpoint therapy and resulting in worse outcomes. By incorporating 9p21 loss, PD-L1 expression, and TMB levels in pre-treatment tumors, a response score was derived that can accurately identify patients likely to achieve sustained response to therapy.
