The potential of long noncoding RNA therapies

Tim R. Mercer et al.

Summary: The vast numbers of long noncoding RNAs (lncRNAs) in the human genome play diverse roles in gene regulation, cell biology, development, and human disease. Advances in RNA-based therapies have enabled the exploration and exploitation of natural lncRNA structures for the rational design and assembly of synthetic RNA therapies. This has the potential to provide insights into the pharmacological properties of lncRNA therapies that have not yet entered clinical trials.

Bone mesenchymal stem cell-derived extracellular vesicles containing NORAD promote osteosarcoma by miR-30c-5p

Hongtao He et al.

Summary: This study discovers that NORAD, a non-coding RNA highly expressed in osteosarcoma cells, can be delivered to osteosarcoma cells through extracellular vesicles (EVs) released by bone mesenchymal stem cells (BMSCs), thereby promoting proliferation and invasion of osteosarcoma. NORAD is found to interact with miR-30c-5p and KLF10, and regulates the miR-30c-5p/KLF10 axis to influence the development and metastasis of osteosarcoma.

Targeting TGF-β signal transduction for fibrosis and cancer therapy

Dandan Peng et al.

Summary: TGF-beta plays essential roles in early embryonic development, organogenesis, immune regulation, tissue repair, and adult homeostasis. Its involvement in fibrosis and cancer is complex and context-dependent, showing both inhibitory and promoting effects. Therapeutics targeting TGF-beta signaling hold promise in treating fibrosis and cancer, although the development is hindered by potential systemic cytotoxicity.

Hypoxia increases the expression of stem cell markers in human osteosarcoma cells

Jinluan Lin et al.

Summary: Hypoxia can induce stem cell-like phenotypes in some osteosarcoma cells, with increased expression of CSC markers and enhanced sphere formation and xenograft tumor growth. Rapamycin, by suppressing HIF-1 alpha protein levels, inhibits the hypoxic response and reduces stem cell marker expression, leading to decreased sphere and xenograft tumor formation efficiency in osteosarcoma cells.
