Adipose extracellular matrix deposition is an indicator of obesity and metabolic disorders

Hui-jian Chen et al.

Summary: Obesity and metabolic disorders pose threats to human health. The extracellular matrix (ECM) in the adipose microenvironment plays an important role, but the specific functions of each ECM component are not fully understood. Glucose and lipid metabolic disorders were observed in mice and humans with high-fat diet (HFD)-induced obesity. Inflammatory factors and hormones were elevated in the serum of HFD-fed mice. Multiple ECM components were upregulated in obese white adipose tissue (WAT), with stronger effects in subcutaneous WAT than visceral WAT in mice, but the fat depot difference was reversed in humans. ECM remodeling and adipocyte morphology changes were observed in HFD-induced obesity. ECM genes in human visceral WAT showed positive correlations with blood lipids and glucose. In vitro experiments revealed the involvement of specific ECM proteins in adipocyte differentiation. Macromolecular crowders (MMC) and oleic acid (OA) had differential effects on ECM protein expression and adipocyte characteristics. Positive correlations were found between ECM genes and adipogenesis-related genes in adipocytes. In conclusion, ECM remodeling induced by obesogens such as HFD is closely related to adipogenesis and can serve as an indicator of obesity and metabolic disorders.

Obesity I: Overview and molecular and biochemical mechanisms

Robert H. Lustig et al.

Summary: Obesity is a chronic, multifactorial condition characterized by excess body fat and its prevalence has been increasing globally. Genetic factors, viruses, insulin resistance, inflammation, gut microbiome, and circadian rhythms all contribute to the development and pathogenesis of obesity. Energy balance regulation relies on the interplay of various hormones and fat cell development is controlled by hormones and growth factors acting via a variety of receptors.

Low leptin levels are associated with elevated physical activity among lean school children in rural Tanzania

Christine Ludwig et al.

Summary: Rapid urbanization and increasing socio-economic status in Sub-Saharan African countries are associated with decreased physical activity. This study provides evidence of a negative association between leptin levels and moderate-to-vigorous physical activity (MVPA) in lean children. It also suggests that children residing in areas with better infrastructure have lower MVPA levels and tend to have higher leptin concentrations.

The prevalence of overweight and obesity has risen during the pandemic, accompanied by a decrease in physical activity and sleep duration, increased screen time, and reduced consumption of sugar-sweetened beverages. Factors such as ethnic minority status, older age, less physical activity, shorter sleep duration, and longer screen time have been positively associated with obesity.

Differences in Classification Standards For the Prevalence of Overweight and Obesity in Children. A Systematic Review and Meta-Analysis

Francisco Llorca-Colomer et al.

Summary: The prevalence of childhood obesity has increased dramatically worldwide. Determining overweight and obesity in children is controversial. This study compared three international classification systems to determine overweight and obesity in infants and adolescents. The results showed that the WHO 2007 criteria had the highest prevalence of overweight and obesity. Most studies found a higher prevalence of childhood overweight and obesity in boys, but when considering the CDC 2000 and Cole-IOTF criteria, girls had a higher prevalence. The review and meta-analysis highlight the need to unify the criteria for childhood overweight and obesity classification. International standards are insufficient, and a working group should be created to establish a gold standard that accounts for geographical region, ethnic groups, age groups, and secular growth.

Sex-dimorphic genetic effects and novel loci for fasting glucose and insulin variability

Summary: Sex differences play a role in genetic effects on fasting glucose and insulin levels, with a sex dimorphism in allelic effects on fasting insulin and sex-homogeneous effects on fasting glucose. In addition, there are stronger genetic correlations between fasting insulin and waist-to-hip ratio/anorexia nervosa in women, and waist-to-hip ratio is causally related to insulin resistance in women but not in men. These findings highlight the importance of understanding metabolic and glycemic health sex dimorphism in exploring genetic effects in related phenotypes between women and men.
