The assembly, regulation and function of the mitochondrial respiratory chain

Irene Vercellino et al.

Summary: The mitochondrial oxidative phosphorylation system is essential for cellular metabolism, driving ATP production through the electron transport chain. Recent studies have provided insights into the organization, assembly, and mechanisms of the respiratory complexes, including the formation of larger assemblies such as respiratory supercomplexes, and their roles in physiology.

Mitochondrial Structure and Bioenergetics in Normal and Disease Conditions

Margherita Protasoni et al.

Summary: Mitochondria are intracellular organelles responsible for energy production in eukaryotic cells, with growing interest due to their association with various pathologies. Dysfunction of mitochondria can lead to a wide range of clinical phenotypes, particularly affecting tissues with high-energy demand. Mitochondrial diseases are genetically heterogeneous conditions, making it difficult to identify common causes and potential therapeutic targets.

Structure and assembly of the mammalian mitochondrial supercomplex CIII2CIV

Irene Vercellino et al.

Summary: The enzymes of the mitochondrial electron transport chain play a vital role in cell metabolism, forming supercomplexes in vivo. The assembly factor SCAF1 is crucial in mediating the assembly of CIII2CIV, but not involved in the assembly of the respirasome. Assembling into supercomplexes enhances the catalytic efficiency of CIII2 and CIV, potentially fine tuning the electron transfer efficiency in the electron transport chain.

Cryo-EM structure and kinetics reveal electron transfer by 2D diffusion of cytochrome c in the yeast III-IV respiratory supercomplex

Agnes Moe et al.

Summary: Energy conversion in aerobic organisms involves electron transfer from low-potential donors to dioxygen through the membrane-bound respiratory chain. The formation of supercomplex CIII2CIV1/2 facilitates electron transfer by two-dimensional diffusion of cyt. c, increasing QH(2):O-2 oxidoreductase activity and suggesting a regulatory role for supercomplex formation in the respiratory chain.
